MicroRNA-Mediated Reprogramming of Somatic Cells into Induced Pluripotent Stem Cells

Abstract: MicroRNAs or miRNAs belong to a class of small noncoding RNAs that play a crucial role in posttranscriptional regulation of gene expression. Nascent miRNAs are expressed as a longer transcript, which are then processed into a smaller 18-23-nucleotide mature miRNAs that bind to the target transcripts and induce cleavage or inhibit translation. MiRNAs therefore represent another key regulator of gene expression in establishing and maintaining unique cellular fate. Several classes of miRNAs have been identified t… Show more

“…Additionally, the miR-302-3p-367 cluster has potential to convert somatic cells into induced pluripotent stem cells (26). A study by Cai et al (20) investigated the role of the miR-302-3p-367 cluster in cervical carcinoma.…”

“…The ESC-specific miR-302 family, which is composed of five members, miR-302a/b/c/d and miR-367, is highly expressed in undifferentiated mESCs, hESCs, and iPSCs, but suppressed in differentiated cells [ 98 ]. MiR-302 regulates the expression of the pluripotency markers Oct4, Sox2, Nanog, and SSEA-3/4 [ 99 ] and stimulates somatic cell reprogramming, which results in the generation of iPSCs through DNA demethylation and by downregulation of Dnmt1 [ 98 ] and methyl-DNA binding domain protein 2 (MBD2) [ 100 ]. In addition, miR-302 facilitates mesenchymal to epithelial transition or MET via targeting of the transforming growth factor β receptor II (TGFBR2) and the Ras homolog gene family member C genes [ 95 ].…”

“…These reprogramming miRs were identified upon a large analysis of miRs expression in mouse ESCs, mouse iPSCs, and adult mouse adipose stromal cells. Using this approach several mouse and human iPSCs lines have been derived [ 49 ]. Because this reprogramming method does not require vector-based gene transfer, it holds significant potential for regenerative medicine.…”
