MicroRNA Expression Profiling of Thyroid Tumors: Biological Significance and Diagnostic Utility

Abstract: In this study, we demonstrate that various histopathological types of thyroid tumors have distinct miRNA profiles, which further differ within the same tumor type, reflecting specific oncogenic mutations. A limited set of miRNAs can be used diagnostically with high accuracy to detect thyroid cancer in the surgical and preoperative FNA samples.

“…The aberrant expression of certain miRNAs can induce initiation and/or progression of human cancers [140]. The involvement of miRNAs in proliferation, differentiation and apoptosis have been defined and thus they could be used as biomarkers for diagnostic and therapeutic targets in many cancers including lung, pancreas, breast, and colorectal cancers in addition to thyroid cancers [141][142][143][144]. There are several reports displaying changes in miRNA profiles in thyroid cancers as compared to normal thyroid tissues [143,144].…”

“…30,31 RNA yield and purity were determined using the NanoDrop 1000 spectophotometer (ThermoScientific, Wilmington, DE, USA). RNA integrity was assessed for 1 ml of sample using the Agilent 6000 NANO kit for the 2100 Bioanalyzer (Agilent Technologies, Santa Clara, CA, USA) according to the manufacturer's protocol.…”

“…miRNA expression levels were calculated by relative quantitation using the ABI 7500 Real-Time PCR SDS 1.2 software (Applied Biosystems), and the fold expression changes were determined by 2-DDC t method. 30 The data were presented as the fold change of miRNA expression in lung tumors relatively to normal lung tissues after normalization to an endogenous control (RNU44).…”

“…[20][21][22] The unique expression profiles of microRNAs characteristic of each tissue and the malignancies arising in them can permit their accurate identification. [23][24][25][26] MicroRNAs are also stable in tissue, stored frozen or as formalinfixed, paraffin-embedded (FFPE) samples, and in serum. [27][28][29] In an earlier work, we showed that a biologically motivated classifier based on expression of a relatively small number of tissue-specific microRNAs allows tumor tissues of origin to be accurately identified.…”
