MicroRNA‐432 contributes to dopamine cocktail and retinoic acid induced differentiation of human neuroblastoma cells by targeting NESTIN and RCOR1 genes

Abstract: Edited by Tamas Dalmay Keywords:MicroRNA-432 NESTIN RCOR1 MECP2 Neurite outgrowth SH-SY5Y cells a b s t r a c t MicroRNA (miRNA) regulates expression of protein coding genes and has been implicated in diverse cellular processes including neuronal differentiation, cell growth and death. To identify the role of miRNA in neuronal differentiation, SH-SY5Y and IMR-32 cells were treated with dopamine cocktail and retinoic acid to induce differentiation. Detection of miRNAs in differentiated cells revealed that expre… Show more

“…25,26 Nestin, as an important stem cell/progenitor cell biomarker, is crucial for the mitogen-stimulated proliferation of neural progenitor cells, and loss or lack of it would lead to apoptosis of neural progenitor cells in zebrafish. 27 Das et al 28 has suggested that miR-432 contributes to dopamine cocktail and retinoic acid-induced differentiation of human neuroblastoma cells by targeting NESTIN and RCOR1 genes, and miR-432 could meditate the process of neuronal differentiation in human neuroblastoma cells, which supports our results. Consequently, we found that compared with the blank and NC groups, frequency of social behaviors of juvenile Wistar rats in the si-C21orf121, miR-140-5p mimic, and si-C21orf121 + miR-140-5p mimic decreased, but accumulated time of repetitive stereotyped movements increased.…”

“…The neuronal cells adhere weakly to the substrate and grow as clumps of cells. For instance, SH-SY5Y, a neuroblastoma cell line, is an in vitro model for investigating the early stages of neuronal differentiation and neural tissue engineering strategies4567. Differentiation of SH-SY5Y induced pharmacologically has also been studied as a model of dopaminergic neurons for Parkinson's disease research8.…”
