The sensitivity and specificity of microRNAs (miRNAs) for diagnosing glioma are controversial. We therefore performed a meta-analysis to systematically identify glioma-associated miRNAs. We initially screened five miRNA microarray datasets to evaluate the differential expression of miRNAs between glioma and normal tissues. We next compared the expression of the miRNAs in different organs and tissues to assess the sensitivity and specificity of the differentially expressed miRNAs in the diagnosis of glioma. Finally, pathway analysis was performed using GeneGO. We identified 27 candidate miRNAs associated with glioma initiation, progression, and patient prognosis. Sensitivity and specificity analysis indicated miR-15a, miR-16, miR-21, miR-23a, and miR-9 were up-regulated, while miR-124 was down-regulated in glioma. Ten signaling pathways showed the strongest association with glioma development and progression: the p53 pathway feedback loops 2, Interleukin signaling pathway, Toll receptor signaling pathway, Parkinson's disease, Notch signaling pathway, Cadherin signaling pathway, Apoptosis signaling pathway, VEGF signaling pathway, Alzheimer disease-amyloid secretase pathway, and the FGF signaling pathway. Our results indicate that the integration of miRNA, gene, and protein expression data can yield valuable biomarkers for glioma diagnosis and treatment. Indeed, six of the miRNAs identified in this study may be useful diagnostic and prognostic biomarkers in glioma.

Background. Although several studies have proved the relationship between the prognostic value of miRNA-15a and different types of cancer, the result remains controversial. Thus, a meta-analysis was conducted to clarify the prognostic value of miRNA-15a expression level in human cancers. Methods. We enrolled appropriate literature by searching the databases of PubMed, Embase, and Web of Science. Subsequently, we extracted HRs and their 95% CIs and calculated pooled results of miRNA-15a for overall survival (OS) and disease-free survival (DFS). Besides, subgroup analysis, sensitivity analysis, and publication bias were also revealed in this study. We also further validated this meta-analysis using the Kaplan-Meier plotter database. Result. 10 studies, including 1616 patients, were embraced in our meta-analysis. The result showed the lower expression of miRNA-15a significantly predicted adverse OS (HR=2.17, 95% CI: 1.41-3.34), but there is no significant association between the expressing level and DFS in cancer patient (HR=2.04, 95% CI: 0.60-6.88). Based on Kaplan-Meier plotter database, we found the same results in bladder Carcinoma, head-neck squamous cell carcinoma, liver hepatocellular carcinoma, lung squamous cell carcinoma, pancreatic ductal adenocarcinoma, rectum adenocarcinoma, stomach adenocarcinoma, and uterine corpus endometrial carcinoma, but opposite results were found in cervical squamous cell carcinoma and esophageal carcinoma. Conclusion. Low expressing levels of miRNA-15a indicated poor OS, while miRNA-15a can be used as a prediction biomarker in different cancer types.

Plasma microRNA-720 may predict prognosis and diagnosis in glioma patients

We enrolled 122 patients with glioma who received surgery treatment in our hospital from June 2010 to May 2012, and 60 healthy individuals. We found that the plasma miR-720 in the glioma group was significantly higher than that in the healthy control group (3.19±1.26 vs 0.98±0.65, P<0.001). The sensitivity and specificity were 71.3% (95%CI: 62.4%-79.1%) and 83.3% (71.5%-91.7%), respectively. The results indicated that the plasma miR-720 level was associated with tumor grade (t=104.418, P<0.001). The advanced tumor tended to have higher miR-720 expression level. No significant association was found between miR-720 and age, sex, tumor size, KPS and tumor position (P=0.438, 0.514, 0.518, 0.058, 0.226). The multivariate cox analysis indicated that the high expression of miR-720 (HR=1.48, 95%CI: 1.12-2.97, P=0.023) was independently predictors of adverse prognosis in patients with glioma. The high expression of miR-720 was also associated with recurrence or development in patients with glioma (HR=1.47, 95%CI: 1.18-3.14, P=0.012). Plasma miR-720 has a moderate diagnostic ability in early diagnostic of glioma and may be a potential tumor biomarker. The high plasma miR-720 was related to adverse prognosis in patients with glioma and could be a prognosis predictor of glioma patients.
