MicroRNA-148a-3p inhibits progression of hepatocelluar carcimoma by repressing SMAD2 expression in an Ago2 dependent manner

Abstract: Background: Hepatocellular carcinoma (HCC) is one of the most prevalent common cancer worldwide with high mortality. Transforming growth factor-β (TGF-β) signaling pathway was reported dysregulated during liver cancer formation and progression. As a key component of TGF-β signaling, the role of SMAD2 and its regulatory mechanisms in HCC remain unclear. Methods: SMAD2 expression in paired HCC specimens were determined by western blot and immunohistochemistry (IHC). quantitative real-time PCR (qRT-PCR) was used … Show more

“…MiRNAs are known to play important roles in the progression of multiple diseases, including liver cancer (17,27). Our research found miR-513a-5p antagomir partially reversed the inhibitory effect of LINC01234 knockdown on liver cancer, further verified the function of miR-513a-5p in liver cancer progression.…”

“…MicroRNAs (miRNAs) are a class of non-coding small ribonucleic acids which can regulate gene expression by suppression of mRNA translation or degradation of mRNAs (16). Moreover, miRNAs have been considered as important biological regulators for multiple diseases, including liver cancer (1,17). Meanwhile, in previous studies, lncRNAs have been reported to mediate the tumorigenesis of liver cancer via sponging miRNAs (15,18).…”

“…Certain miRNAs such as miR-21, miR-221, and miR-222 are frequently overexpressed in HCC [55][56][57][58][59][60][61][62]. These miRNAs have been demonstrated to promote oncogenesis by negatively regulating important tumor-suppressive protein-coding genes in HCC [63][64][65][66][67]. On the other hand, miRNAs such as miR-122, miR-125b, miR-139, miR-101, and let-7 are recurrently downregulated in HCC [68][69][70][71][72].…”

“…Induction of miR-148a expression in breast cancer cell line was reported to inhibit cellular proliferation and migration [ 29 ] Furthermore, over expression of miR-148a led to inhibition of breast cancer cell proliferation with an elevation in apoptosis through targeting B-cell lymphoma, that previously had established in breast cancer cells [ 25 ]. In addition, elevated miR-148a level suppresses migration and invasion in hepatocellular carcinoma and, knocking down miR-148a has the reverse effect [ 30 ].…”
