MicroRNA 146a Polymorphisms and Expression in Indian Children with Acute Lymphoblastic Leukemia

Abstract: Background MicroRNAs (miR) have been reported to be involved in hematopoiesis and in the pathogenesis of several hematological malignant neoplasms. Single-nucleotide polymorphisms (SNPs) in human miR genes may alter the expression of those genes and influence the predisposition to childhood leukemia. Objective To evaluate the association of rs2910164 G>C, rs57095329 A>G and the expression of miRNA-146a in ethnic South A… Show more

“…In 2014, Hassani et al reported that mir-146a rs2910164 was associated with pediatric ALL risk (11). However, another study found no relationship between mir-146a and ALL risk (12). Gutierrez et al found that both mir-3117 rs12402181 and mir-3689d2 rs62571442 increased B-ALL risk (13).…”

“…So far, most of the causes of ALL are undeciphered; however, it is well known that an interaction within environmental and genetic factors is needed to develop this malignancy (4)(5)(6). Among the identified risk genetic factors to suffer ALL are the single nucleotide polymorphisms (SNP), both, in coding and no coding genes (6)(7)(8)(9). No coding genes comprises around 98% of the human-transcribed genome, which is mainly represented by microRNAs (miRNAs) and long noncoding RNAs (lncRNAs) that play a relevant role in LLA and other types of cancer (10).…”

“…(16,17). The rs2910164 has been associated with many types of cancer and several immune-mediated diseases (18)(19)(20); however, its association with ALL has shown controversial results (9,17,21). Another functional miR-SNP is rs3746444, which results from an A-to-G substitution in the seed region of miR-499a, was reported as significantly associated with an increased susceptibility to several human conditions, including cancer (19,22).…”

“…In 2016, Chansing and his colleagues found that the frequencies of miR146a rs2910164 genotypes were not associated with demographic data or clinical outcomes for ALL in a Thailand population containing 100 childhood ALL cases and 200 healthy controls (32). In 2019, one group reported that no statistically significant association was found between miR146a rs2910164 genotypes and childhood ALL risk in an Indian population containing 71 cases and 74 control individuals (33). Compared to these studies, our sample size is much more convincing with 266 cases and 266 controls, and representative in an islandic country with conservative genetic background.…”
