Rapid clonal micropropagation protocol of Aegle marmelos (L.) Corr. cv. CISH-B1 was achieved by nodal stem segment of mature bearing tree. Three centimeter long shoots having one axillary bud excised from 10-15th nodal region of shoots during September gave quick in vitro bud burst (5.33 days) when cultured on MS medium supplemented with BAP, 8.84 μM + IAA 5.7 μM. The maximum number of proliferated shoots (9.0/explant) were obtained on same medium supplemented with BAP 8.84 μM + IAA 5.7 μM. The micro shoots were rooted (100 %) on ½ strength MS medium supplemented with IBA 49.0 + IAA 5.7 μM. In vitro rooted plants were acclimatized on autoclaved coconut husk containing ½ strength MS plant salt mixture and under shade net house (50 % shade 70-80 % RH). The plants were established in the field after acclimatization. The micropropagated plants were tested for its genetic fidelity using 13 RAPD, 3 ISSR and 2 DAMD primers. Profile obtained by all the three Single Primer Amplification Reaction (SPAR) technique from mother tree and micropropagated plants revealed genetic integrity of micropropagated plants with that of mother tree.

La propagación in vitro mediante organogénesis directa de brotes de guayabo (Psidium guajava L.) constituye una alternativa para la rápida obtención de plantas productivas. El presente trabajo tuvo como objetivo optimizar un protocolo de regeneración in vitro a partir de ápices y segmentos nodales de cinco genotipos de guayabo con alto potencial comercial Paluma 12 (PM12), Paluma 6 (PM6), 17-06, Roja Exterior Redonda (RER) y Roja Chapeada Grande (CHRG). Se evaluó la adición de polivinilpirrolidona (PVPP) en el medio de cultivo como agente antioxidante. Los genotipos RER y CHRG presentaron menor número de brotes, con 10 y 0 % de fenolización. En la fase de multiplicación de propágulos los mejores resultados fueron obtenidos cuando los brotes se desarrollaron en el medio Murashige y Skoog (MS) con el suplemento de 0.5 mg L-1 de BAP y con 30 mg L-1 de adenina para los genotipos PM6 y PM12, mientras que el genotipo 17-06 tuvo mejor desarrollo con la adición de 15 mg L-1 de sulfato de adenina. La adición de 40 mg L-1 de sulfato de adenina representó el mejor tratamiento para los genotipos RER y CHRG. El enraizamiento se indujo satisfactoriamente mediante la incubación de los brotes durante 24 h en medio MS suplementado con 2.54 mg L-1 de AIA para el genotipo 17-06, con 1.34 mg L-1 de AIB para los genotipos PM6 y RER, y con 2.94 de AIB para los genotipos CHRG y PM12.

The research was conducted for the assessment of genetic diversity using both morphological and random amplified polymorphic DNA (RAPD) analysis of twelve guava (Psidium guajava L.) varieties growing in Bangladesh. Morphological characterization of guava varieties showed a wide range of variation. The highest variability was observed between Poly and Jelly varieties.Polymerase chain reaction with 5 arbitrary 10-mer and 3 arbitrary 12- mer RAPD primers produced a total of 50 bands of which 75.23 percent were polymorphic. The highest percentage of polymorphic loci (100%) was observed for primer A and the lowest (50%) for A03 primer. The UPGMA dendrogram revealed the segregation pattern and the difference of evolutionary changes. Guava varieties were separated into two main groups, one of them was made up of Chineese, Jelly, Kazi, Apple, L-49, Local-2 and Local-3. The other one was made up of Local-1, Poly, Kashi, Thai and Bombay. The highest genetic distance between Apple and Kazi peyara indicate that these varieties might be interesting in breeding programme for improving trait of interest.
