Micropropagation of caari under different nutritive culture media, antioxidants, and levels of agar and pH

Abstract: The caçari (Myrciaria dubia) is a native fruit tree from Amazon with high concentrations of vitamin C. This study aimed to adjust a culture medium that meets the nutritional needs for the in vitro development of caçari, evaluating the effect of different concentrations and nutritive culture media, antioxidant, and levels of agar and pH. Three experiments were carried out in a completely randomized design: 1-woody plant medium (WPM), Murashige and Skoog (MS) and Juan, Antonio, Diva and Silvia medium (JADS) nutr… Show more

“…Subsequently, the explants were immersed in a fungicide solution with 2 ml L -1 Nativo® and 100 mg L -1 of the antioxidant citric acid, as recommended by Araújo et al (2016), remaining under these conditions for 2 h. After this step, in a laminar flow chamber, the stem segments underwent a disinfection process as described in the following sequence: the explants were immersed in 70% alcohol for 1 min; soon after, they were immersed in 1.5% sodium hypochlorite with 2 drops of neutral detergent for 10 min; at the end, the disinfected explants underwent triple washing with distilled, deionized and autoclave water (DDA) to completely remove the products from the surface of the tissues.…”

“…In the trial with cajú do cerrado, the MS (50% and 25%) and WPM (100% and 50%) media were the most efficient in plant regeneration (ASSIS et al, 2012). Meanwhile, in caçari, 100% WPM was superior to the MS and JADS medium at concentrations of 25%, 50%, 75%, and 100% (ARAÚJO et al, 2016). Notably, for some woody plants, the higher concentration of nitrogen salts has a negative effect on in vitro development.…”
