Micropatterned Neurovascular Interface to Mimic the Blood–Brain Barrier’s Neurophysiology and Micromechanical Function: A BBB-on-CHIP Model

Abstract: A hybrid blood–brain barrier (BBB)-on-chip cell culture device is proposed in this study by integrating microcontact printing and perfusion co-culture to facilitate the study of BBB function under high biological fidelity. This is achieved by crosslinking brain extracellular matrix (ECM) proteins to the transwell membrane at the luminal surface and adapting inlet–outlet perfusion on the porous transwell wall. While investigating the anatomical hallmarks of the BBB, tight junction proteins revealed tortuous zon… Show more

“…This paves the way to design candidate genomic screening and personalized timing to optimize dose administration in any vaccination program. Moreover, advanced 3D cell models based on integrated circuits and systems as chip technology may help and serve as an additional in vitro surrogate to summarize the complex response of any cell/organ target for the virus recapitulating the native cellular environments under higher biological fidelity ( Singh et al, 2022 ). This will allow a more efficient and personalized approach for vaccine selection, formulation, timing, and scheduling of the booster doses.…”

“…The high-content image-based screening (HCIBS) is a type of phenotypic drug discovery approach where biological images are used to analyze cells and tissues specific compounds or information (Singh et al 2022a ). HCIBS data typically include images of cells or tissues, as well as quantitative data on the various cellular parameters being measured (Fig.…”

“…Although there is a gap in the translation of nanomedicine, the use of computer-aided analysis has become evident in this area. This has led researchers get a clear vision on the behaviour and application of nanoparticle- based therapy in reproductive biology ( 79 ), transporting drugs across biological barriers like the blood-brain barrier ( 80 , 81 ). Another important aspect with respect to the design and use of nanotherapeutics is the toxicity; the understanding of which has been highly enhanced using computational biology ( 82 ).…”
