Micronucleus Test of Picrorrhiza Rhizoma Aqueous Extract in Bone Marrow Cells of Male ICR Mice

Abstract: In this research, the genotoxic effect of Picrorrhiza Rhizoma (PR) aqueous extract was evaluated using the mouse micronucleus test. PR extract was administered once a day for 2 continuous days by oral gavage to male ICR mice at doses of 2000, 1000 and 500 mg/kg. Cyclophosphamide was used as a known genotoxic agent in a positive control. The appearance of a micronucleus (MN) in polychromatic erythrocyte (PCE) is used as an index for genotoxic potential, and PCE ratio is used as an index of cytotoxicity. Althoug… Show more

“…In particular, 100 mg/kg of AR showed anti-mutagenicity by CPA, that is, reduction in genotoxicity, comparable to that of 200 mg/kg of EAP. In contrast, the ratio of PCE (PCE/ (PCE + NCE)) is used as an indicator of the toxicity of test substances to bone marrow cells [50]. The results of this experiment confirmed that the number and ratio of PCE decreased in the CPA control group.…”

“…In the present study, mitomycin C used as a positive control, and it showed a significant increases of MNPCE ratios. This indicates that the experimental design and the results of the present study are acceptable [19]. The sensitivity, specificity and concordances of the Ames test are known to be 58.8, 73.9 or 62.5%, respectively [20].…”

“…The micronucleus test is used to indicate the potential of genotoxic agents to influence chromosomal structure or destroy the mitotic spindle that controls chromosome number. An increase in the frequency of MNPCEs is regarded as a positive result of induced chromosomal damage ( 44 ). When DNA fragments or whole chromosomes are not incorporated into the main nuclei during mitosis, micronuclei are formed as small satellite structure nuclei ( 40 ).…”
