Microfluidic Paper‐Based Analytical Device for Sensitive Detection of Peptides Based on Specific Recognition of Aptamer and Amplification Strategy of Hybridization Chain Reaction

Abstract: In this paper, a microfluidic paper-based analytical device is presented for sensitively detecting peptides based on the specific recognition of aptamer and the amplification strategy of hybridization chain reaction (HCR). The aptamer/initiation strand (AP/IS) was designed for the specific target recognition, and the presence of the target peptide could release the initiating sequence from the AP/IS. The released IS could trigger the HCR between two hairpin probes, which could complete the detection for the ta… Show more

“…Luminescence, as a general term is related to the energy transition between molecular orbitals that produces an emission of light. When the excitation of the molecules is caused by a chemical reaction, this light emission is chemiluminescence [ 27 , 33 , 36 , 58 , 71 , 81 ]. The emission that accompanies an electrochemical reaction is known as electrochemiluminescence [ 79 ].…”

“…Origami paper analytical devices (oPADs) have been introduced by several groups [ 79 , 80 , 81 ]. For example, Liu et al [ 80 ] used a glucose oxidase tag to modify the relative concentrations of an electroactive redox couple, and a digital multimeter (DMM) to transduce the result.…”

“…ECL is a powerful analytical technique which is gaining increased attention for the development of analytical methods using µPADs [79]. One example is the ECL-based µPAD developed by Ma et al [101]. This work uses the specific recognition by aptamers and the amplification strategy of a hybridization chain reaction (HCR) with the ECL probe, Ru(phen)3 2+ for detecting the analyte peptides.…”
