Microemulgel: an overwhelming approach to improve therapeutic action of drug moiety

A huge amount of Indian medicinal plants are accredited with various pharmacological activities. Extract of medicinal plant and extracted oil have more or less pharmacological property, some natural penetration enhancers have advantages such as low cost, better safety profile hence they increase their acceptances in the formulation of microemulgel. Some of oil may assist the therapeutic action of API. It is convenient, cost-effective to use of herbal oil as oil phase or API, penetration enhancer, surfactant or other constituent in microemulgel formulation. Microemulgel is a microemulsion and gel combination with herbal ingredient, with small sized globule present in an emulsion. Microemulsion consists of delivering a drug dissolved in a mixture of one or more excipient like mono, di, and triglycerides, lipophilic and hydrophilic co surfactant. Microemulsion is formed when entropy changes in that dispersion is greater than the free energy require increasing the surface area between oil and aqueous phase of dispersion. Microemulgel has a dual action of micro-emulsion and gel. Emulsion based gel, the drug get entrapped in cross linkage network of gelling agent. In that small drug get entrapped and release in controlled manner. Microemulgel have other advantages like good uniformity, easily spreadable, greaseless as well as bio-friendly, detachable, non-staining, emollient, longer shelf-life, transparent, pleasant appearance, the ability of patients for self-medication, termination of medications will be easy.

The objective of the present study is to formulate and evaluate ofloxacin emulgel. Emulgel formulations of ofloxacin were prepared using different concentrations of gelling agent’s Carbopol-940 and Xanthum gum. Tween-80 and span-80 were used as emulsifiers and propylene glycol as a humectant in the preparation of emulgel. The effect of the concentration of gelling agent on the drug release from the prepared emulgel was investigated. The compatibility study was conducted using Fourier-transform infrared (FTIR). The formulated emulgel was characterized by their physical appearance, pH determination, viscosity, spreadability, drug content, microbial test and in vitro diffusion study. FTIR indicated that the drug and excipients used in the study are compatible with each other. All the prepared formulations showed acceptable physical properties, homogeneity, consistency, spreadability, viscosity, and pH value. Drug release from all the formulations depended upon the concentration of the polymer used. As the concentration of Carbopol 940 increased the spreadability and drug release was found to be decreased. Emulgels formulated with oleic acid gave a much higher release rate of ofloxacin than emulgels formulated with liquid paraffin. The release of drug from all the emulgels prepared followed Zero-order kinetics. The linear Higuchi plots indicated that the drug release from all the emulgels prepared followed diffusion kinetics. Emulgel formulated with oleic acid exhibited greater flux when compared with those formulated with liquid paraffin. The formulations were found to be stable during stability testing. It can be concluded that Carbopol 940 and oleic acid are recommended for the formulation and preparation of Ofloxacin emulgels for topical drug delivery. Carbamazepine(CBZ) is a synthetic compound of the benzodiazepine class, used as an anticonvulsant and analgesic drug. Carbamazepine is relatively slowly but well absorbed after oral administration. Its plasma half-life is about 30 hours when it is given as single dose, but it is a strong inducer of hepatic enzymes and the plasma half-life shortens to about 15 hours when it is given repeatedly. Psoriasis is autoimmune disease in which genetic and environmental factors have a significant role. Carbamazepine was accidently discovered to have antipsoriatic properties. Carbamazepine was obtained as a gift sample from Lupin. Characterization of carbamazepine was carried out by melting point and IR. Carbamazepine has limited aqueous solubility so in literature Self microemulsifying drug delivery system (SMEDDS) have been reported. In the present study, a topical microemulsion based gel of Carbamazepine was prepared. The pseudo ternary phase diagram was prepared to find the microemulsion region. Various microemulsion gel formulations were prepared using carbopol-974 and poloxamer-407 in varied concentrations. Prepared formulations (FG1 to FG10) were evaluated for various parameters like color, appearance, consistency, pH, spreadability and IR. The microemulsion based gels were found to have good spreadability and pH within the range of skin pH and thus suitable to use on skin.
