Microarray profiling analysis and validation of novel long noncoding RNAs and mRNAs as potential biomarkers and their functions in atherosclerosis

Abstract: Long noncoding (lnc)RNAs have been implicated in the development and progression of atherosclerosis. However, the expression and mechanism of action of lncRNAs in atherosclerosis are still unclear. We implemented microarray analysis in human advanced atherosclerotic plaques and normal arterial intimae to detect the lncRNA and mRNA expression profile. Gene Ontology functional enrichment and pathway analyses were applied to explore the potential functions and pathways involved in the pathogenesis of atherosclero… Show more

“…Therefore, they were selected for reverse transcription-quantitative PCR (RT-qPCR) verification. In addition, according to previous studies, MYH11 plays a regulatory role in cardiac function 26 , 27 . Furthermore, MYH11 was highly correlated with TPM1 and MYL2 in the PPI network of WS/WF and WF/WC.…”

“…Recent years lncRNAs have been implicated as potential biomarkers in the progression of antherosclerosis 25‐27 . For instance, lncRNA MIAT aggravates atherosclerotic damage via activation PI3K/Akt signalling pathway, 10 LncRNA MANTIS played an crucial role in the beneficial effects of stain treatment and laminar flow 28 ; LncRNA CDKN2B‐AS1 decreased inflammatory response and promoted cholesterol efflux through inhibited the transcription of ADAM10 29 .…”

“…Likewise, studiesby Okada et alconfirmed that the plasma level of BDNF in myocardial ischemicmice increased significantly, which puts forward the central nervous system-mediated signal transduction mechanism to protect ischemic myocardium: when there is cardiac ischemic damage, myocardial cells through efferent nerve feedback to the central nervous system increase the expression of BDNF in the nerve cells, so as to increase the level of BDNF in peripheral blood, activating theangiogenesis-related molecular mechanism of inhibiting myocardial reconstruction and repair of ischemic myocardium [ 20 ]. Recently, a new study also uncovered serum BDNF was differentially expressed in plaque tissues in patients with coronary heart disease compared with normal intimae in healthy subjects using microarray profiling analysis and qRT-PCR validation [ 21 ]. Recent studies have revealed that BDNF regulates cardiomyocyte contraction and directly affects cardiac function of mice [ 22 , 23 ].…”
