Metals in Medicine

Abstract: Not only the 24 or so essential elements, but also nonessential and even radioactive elements have enormous potential for applications in medicine. In the fight against cancer cisplatin, one of the world's best selling anticancer drugs, is being joined by other platinum, titanium, and ruthenium complexes. Gadolinium(III) complexes can be safely injected as magnetic resonance imaging contrast agents, and ligand design allows targeting of paramagnetic ions as well as radiodiagnostic (e.g. Tc) and radiotherapeuti… Show more

“…The data were fitted to Equation 2 with the software Scientist ® (Micromath) using a standard least square procedure. (2) The relative error for the fitting of the absorbance vs. time curves in all cases was lower than 1 % and the calculated first order rate constants k obs and k p values were reproduced within a 5 % error as determined in 5 identical experiments.…”

“…These properties can all exert a critical influence on the biological activity and corresponding mechanisms of action [4,5]. For example, some studies have shown that some metal ions generate reactive-oxygen-species (ROS) [6], which although being an inevitable byproduct of cellular respiration, can result in cellular and subcellular oxidative stress and in an altered homeostasis state.…”

“…Hence, it is of great importance to understand in detail the DNA binding properties of novel metal complexes and their possible relationship to cytotoxicity in tumor cells since this may provide grounds for establishing new structure-pharmacological activity relationships. In the studies presented herein, we demonstrate that the antitumor mechanism of mononuclear Cu 2+ , Co 2+ , Zn 2+ , and Ni 2+ complexes of the biologically active ligand clotrimazole (clotri) of the forms [M(clotri)2Cl2]·nH2O (1)(2)(3)(4), [M(clotri)2Br2]·nH2O (5,6), and [M(clotri)3NO3]NO3·nH2O (9,11) and [M(clotri)3(NO3)2] (10) previously studied by us [17], involves not only oxidative DNA damage with generation of ROS but also specific DNA interactions.…”

“…Segundo Crasciunesco e cal.
