SummaryRiboflavin (RF), also known as vitamin B2, belongs to the class of water-soluble vitamins and is widely present in a variety of food products. It is sensitive to light and high temperature, and therefore, needs a consideration of these factors for its stability in food products and pharmaceutical preparations. A number of other factors have also been identified that affect the stability of RF. These factors include radiation source, its intensity and wavelength, pH, presence of oxygen, buffer concentration and ionic strength, solvent polarity and viscosity, and use of stabilizers and complexing agents. A detailed review of the literature in this field has been made and all those factors that affect the photo, thermal and chemical degradation of RF have been discussed. RF undergoes degradation through several mechanisms and an understanding of the mode of photo- and thermal degradation of RF may help in the stabilization of the vitamin. A general scheme for the photodegradation of RF is presented.

Surface-enhanced Raman spectroscopy (SERS) has been suggested as a powerful tool to identify bacteria, drawing from its high fingerprint (vibrational) information content, its extreme sensitivity (down to the single molecule level) and its obliviousness to the aqueous environment intrinsic to biological systems. We review here in a comparative manner the various studies that attempted to utilize SERS for this important goal in light of the work carried out by our own group over the past 10 years or so. We show that SERS has an additional major advantage, namely, it introduces a new dimension of selectivity, which, on the one hand, makes it even more suitable as an analytical tool, but on the other hand, it requires gaining control of the precise manner in which the SERS-active metal centers are produced and brought into contact with the micro-organism. Our emphasis in this review is on understanding the spectra in terms of the nature of the SERS-active centers and their placement within the bacterium. On the interpretation and assignment of the spectra, we constantly keep in mind the final goal of bacteria identification.

Surface-enhanced Raman spectroscopy (SERS) of bacteria infused with silver or gold colloids reflects the different biochemicals in the immediate, nano-vicinity of the particles. In particular we demonstrate high sensitivity to flavins, important coenzymes usually associated with the cell wall. Varying the type of colloid, the laser excitation wavelength and the chemical protocols enables us to observe spectral signatures from different biochemical components of the microorganism. Specifically, we can switch between normal Raman, resonance Raman, and SERS, altering the relative spectral contributions of the various cell constituents.
