Metabolomics reveals the protective of Dihydromyricetin on glucose homeostasis by enhancing insulin sensitivity

Abstract: Dihydromyricetin (DMY), an important flavanone found in Ampelopsis grossedentata, possesses antioxidative properties that ameliorate skeletal muscle insulin sensitivity and exert a hepatoprotective effect. However, little is known about the effects of DMY in the context of high-fat diet (HFD)-induced hepatic insulin resistance. Male Sprague-Dawley(SD) rats were fed a HFD(60% fat) supplemented with DMY for 8 weeks. The administration of DMY to the rats with HFD-induced insulin resistance reduces hyperglycemia, … Show more

“…Metabolites were extracted with cold 80% methanol in the presence of glass beads by a mini‐beadbeater (Biospec Products, https://www.biospec.com). Metabolite profiling was performed by the Metabolomics Facility at Technology Center for Protein Sciences, Tsinghua University, using a TSQ Quantiva Triple Quadrupole liquid chromatography–mass spectrometry (Thermo Scientific, San Jose, CA, USA) according to the previously described method (Le et al ., ). Metabolites were separated on a 100 × 3 mm Synergi™ Hydro‐RP 100A column (Phenomenex, http://www.phenomenex.com/) at 35°C with a linear gradient from 95% mobile solvent A (10 m m tributylamine and 15 m m acetate in water) to 90% mobile solvent B (100% methanol) over a 25‐min period.…”

“…Besides, the anticancer (Zhang et al., ), cardioprotective (Liu, Ai, Feng, Li, and & Liu ), and hepatoprotective effects (Chen et al., ; Qiu et al., ) were proved both in vitro and in vivo . The effect of enhancing insulin sensitivity was demonstrated in high‐fat diet (HFD)‐fed rat model (Le et al., ). The effective dose of the above pharmacological effects was from 50 to 500 mg/kg in vivo .…”

“…Dihydromyricetin (DMY), a type of flavonoid, is the main bioactive component of Ampelopsis grossedentata, which is a widely distributed herbal medicine in Southern China (Okazaki et al, 2005). Previous reports demonstrated that DMY has numerous biologic and pharmacological activities, including antioxidative, anti-inflammatory, antiapoptotic, anticarcinogenic, and antidiabetic effects Li et al, 2015;Ye et al, 2015;Le et al, 2016;Wu et al, 2017). Considering that DMY possesses an anti-inflammatory property, we investigated its potential capacity to inhibit the chronic autoimmune inflammation of RA.…”
