Metabolomic study of Chilean biomining bacteria Acidithiobacillus ferrooxidans strain Wenelen and Acidithiobacillus thiooxidans strain Licanantay

Abstract: In this study, we present the first metabolic profiles for two bioleaching bacteria using capillary electrophoresis coupled with mass spectrometry. The bacteria, Acidithiobacillus ferrooxidans strain Wenelen (DSM 16786) and Acidithiobacillus thiooxidans strain Licanantay (DSM 17318), were sampled at different growth phases and on different substrates: the former was grown with iron and sulfur, and the latter with sulfur and chalcopyrite. Metabolic profiles were scored from planktonic and sessile states. Spermi… Show more

“…Targeted and untargeted metabolomics approaches (cf. Patti, Yanes, & Siuzdak, ) have been used to gain information about the metabolome of plants, phytoplankton, and heterotrophic bacteria (Fiore, Longnecker, Kido Soule, & Kujawinski, ; Longnecker, Kido Soule, & Kujawinski, ; Sawada et al., ), while studies of chemolithoautotrophic bacteria are so far limited to acidophilic bacteria used in biomining applications (Martínez et al., ). In this study, we have used Sulfurimonas denitrificans as a representative model organism to investigate the first metabolome of widely distributed, neutrophilic chemolithoautotrophic Campylobacteria , complementing and verifying biochemical pathways inferred from the genome and to assess the adaptations of S. denitrificans to osmotic stress.…”

“…Genomics has been of great importance to define the biodiversity of biomining processes and has allowed developing both conceptual and metabolic models for some of the acidophilic microorganisms involved. By using metabolomics, some specific metabolites may be used as markers to follow the evolution in time of industrial bioleaching operations (Mart ınez et al, 2013). On the other hand, proteomics has been useful to study microorganisms-mineral interactions and biofilm formation (Mart ınez et al, 2015), including copper resistance as described in the next section.…”

“…Bioleaching bacteria, Acidithiobacillus ferrooxidans and thiooxidans strains were sampled at different growth phases and on different substrates. Metabolic profiles were scored from planktonic and sessile states, using CE/MS . Spermidine was detected in intra‐ and extracellular samples for both strains, suggesting it has an important role in biofilm formation in the presence of solid substrate.…”
