In this study, the metabolite profiling of three different parts of Crocus sativus L. was measured by using ultra‐performance liquid chromatography‐quadrupole time‐of‐flight mass spectrometry (UPLC‐QTof‐MS/MS). Multivariate statistical analysis was used to distinguish among the samples from different parts. A total of 54 compounds were identified in tepals, stigmas and stamens by UPLC‐QTof‐MS/MS. The results stated that chemical characteristics of saffron were obviously diverse in terms of the parts of flower. Through analysis, coniferin and crocin‐2 were special components in stigmas when compared to tepals and stamens. The content of flavonoids was high in tepals when compared with the stigmas. The tepal of saffron may processed as a source of flavonoids in the future. The research provided the basis for the theory that only the stigma can be used as medicine.

Untargeted Metabolite Profiling of Adipose Tissue in Hyperlipidemia Rats Exposed to Hawthorn Ethanol Extracts

The study aimed to explore the metabolic changes of adipose tissue of hyperlipidemia rats with hawthorn ethanol extracts (HEE) consumption by a high‐throughput metabolomics approach. HEE were mainly composed of chlorogenic acid, hyperoside, isoquercitrin, rutin, vitexin, quercetin, and apigenin by HPLC analysis. HEE administration significantly lowered levels of the total cholesterols, triglyceride and low‐density lipoprotein cholesterol as compared to the high‐fat diet model. Gas chromatography‐mass spectrometry was used to identify adipose metabolite profiles. Numerous endogenous molecules were altered by high‐fat diet and restored following intervention of HEE. Metabolites elevated in adipose, including l‐threonine, aspartic acid, glutamine, mannose, inositol and oleic acid, were detected after HEE consumption. Fifteen metabolites were identified as potential biomarkers of hyperlipidemia. Pathway analysis showed that most of the discriminant metabolites were included in fatty acid biosynthesis, galactose metabolism, biosynthesis of unsaturated fatty acids, arginine and proline metabolism, alanine, aspartate and glutamate metabolism, glycerolipid metabolism and steroid biosynthesis. These metabolites and metabolic networks we found offer new insights into exploring the molecular mechanisms of lipid‐lowering of hawthorn ethanol extracts on adipose tissue of rats. Practical Application There was a very high proportion of hyperlipidemia in China. Hawthorn is attracting increasing attention owing to their health benefits, low toxicity, effectiveness and might be suitable for long‐term use.

Intake of arsenic (As) via drinking water has been a serious threat to global public health. Though there are numerous reports of As neurotoxicity, its pathogenesis mechanisms remain vague especially its chronic effects on metabolic network. Hippocampus is a renowned area in relation to learning and memory, whilst recently, cerebellum is argued to be involved with process of cognition. Therefore, the study aimed to explore metabolomics alternations in these two areas after chronic As exposure, with the purpose of further illustrating details of As neurotoxicity.Twelve 3-week-old male C57BL/6J mice were divided into two groups, receiving deionized drinking water (control group) or 50 mg/L of sodium arsenite (via drinking water) for 24 weeks.Learning and memory abilities were tested by Morris water maze (MWM) test. Pathological and morphological changes of hippocampus and cerebellum were captured via transmission electron microscopy (TEM). Metabolic alterations were analyzed by gas chromatography-mass spectrometry (GC-MS). MWM test confirmed impairments of learning and memory abilities of mice after chronic As exposure. Metabolomics identifications indicated that tyrosine increased and aspartic acid (Asp) decreased simultaneously in both hippocampus and cerebellum. Intermediates (succinic acid) and indirect involved components of tricarboxylic acid cycle (proline, cysteine, and alanine) were found declined in cerebellum, indicating disordered energy metabolism. Our findings suggest that these metabolite alterations are related to As-induced disorders of amino acids and energy metabolism, which might therefore, play an important part in mechanisms of As neurotoxicity.Ting-Ting Mo, Hua Dai, and Hang Du contributed equally to the study.
