SUMMARY1. Microdialysis was used to gain insight into the substrate exchanges in the interstitial space of skeletal muscle and adipose tissue. Probes were inserted in the quadriceps femoris muscle and para-umbilical subcutaneous adipose tissue of thirteen subjects and microdialysis was performed at different flow rates (1-4 ,l min-') and during changes in tissue blood flow.2. When ethanol (5 mM) is included in the perfusion solution, the ethanol clearance from the probe is a measure of tissue blood flow. Blood flow changes induced by adenosine or vasopressin perfusion, by exercise or by circulatory occlusion resulted in ethanol clearance values of 69-139 % of the basal level. The ethanol clearance was higher in skeletal muscle than in adipose tissue (32-62 %, P< 0001), a difference compatible with a higher blood flow in muscle tissue.3. The fraction of the interstitial glucose concentration that was recovered with the microdialysis was similar in skeletal muscle (the absolute values being 1'70 + 0 14 mm at 1 #sl min-' and 059 + 005 mm at 4 #1 min-') and adipose tissue (1I89 + 020 mm at 1 pul min-'; 054 + 0 05 mm at 4 #1 min-') and correlated inversely with the tissue ethanol clearance, both in the basal state and during changes in tissue blood flow (muscle: r = -056 to -067; adipose tissue r = -072 to -095). Coefficients of variation were 6-8 % (glucose) and 11-16 % (lactate) and were similar during isometric exercise. The reproducibility of the technique (comparison of two contralateral probes; perfusion flow rate 4 ,a1 min-') was 5'3-8'3 % (ethanol) and 23-9-208 % (glucose) in muscle (n = 6) and adipose tissue (n = 4) respectively.4. The skeletal muscle dialysate lactate concentration (1 #1 min-': 1 16 + 0 2 mM) was higher than in adipose tissue (076 + 008 mm, P < 005), where the absolute amount of lactate that could be removed from the tissue (at 4 1I min-') was only half of that in skeletal muscle (0O8 + 0-11 vs. 1-76 + 023 nmol min-', P< 005

Although humans vary in their response to chemicals, comprehensive measures of susceptibility have generally not been incorporated into human risk assessment. The U.S. EPA dose-response-based risk assessments for cancer and the RfD/RfC (reference dose-reference concentration) approach for noncancer risk assessments are assumed to protect vulnerable human subgroups. However, these approaches generally rely on default assumptions and do not consider the specific biological basis for potential susceptibility to a given toxicant. In an effort to focus more explicitly on this issue, this article addresses biological factors that may affect human variability and susceptibility to trichloroethylene (TCE), a widely used halogenated industrial solvent. In response to Executive Order 13045, which requires federal agencies to make protection of children a high priority in implementing their policies and to take special risks to children into account when developing standards, this article examines factors that may affect risk of exposure to TCE in children. The influence of genetics, sex, altered health state, coexposure to alcohol, and enzyme induction on TCE toxicity are also examined.

This study was conducted to investigate the detoxification effect of extract from soy-sprout grown using high concentrated oxygen water extract (SE) against alcohol-induced hangover in male Sprague-Dawley (SD) rats. The rats were orally administered with different concentrations of SE beverage [26, 260 and 2,000 mg/kg body weight (b.w.)] and, after 30 min, with alcohol at a dose of 3 g/kg b.w. After 1 and 3 h of alcohol administration, blood was collected from the retroorbital plexus, while after 5 h, blood was collected from the heart. In the 2,000 mg/kg b.w. SE group, the concentration of blood alcohol was significantly reduced after 1-5 h of alcohol loading as compared with that in the other groups. In addition, the blood acetaldehyde concentration was reduced by SE (2,000 mg/kg b.w.). These results suggest that SE beverage can alleviate alcohol hangover symptoms by stimulating the activities related to hepatic alcohol-metabolizing enzymes.
