Metabolism and Menstrual Cycle Rhythmicity of Serotonin in Primary Headaches

Abstract: We investigated the platelet and plasma levels of serotonin and its metabolite, 5-hydroxyindoleacetic acid, in patients suffering from episodic tension-type headache and migraine with and without aura, during a headache-free period. In female subjects, blood samples were drawn during the follicular, ovulatory, and late luteal phases of the menstrual cycle. In tension headache and migraine with aura, the group mean values of serotonin and 5-hydroxyindoleacetic acid in platelets and plasma were significantly inc… Show more

“…The main product of tryptophan hydroxylase is serotonin (5-HT), whereas tryptamine is the neuromodulator that derives from the decarboxylase enzyme activity [47]. Numerous studies have been done, in the last decades, to explore possible anomalies of this in indole in migraine patients with very few compelling results.…”

“…Changes in serotonin content were also noticed in women with menstrual migraine, in relation to the cyclic hormonal variations. In these women, platelet serotonin levels peaked in the follicular period and decreased in the luteal phase, when 5HIAA values were particularly increased [16,17]. These data suggest an aberration in menstrual cycle-related changes of serotonin in women migraineurs, possibly due to increased catabolism or reduced synthesis which predisposes these patients to migraine attacks.…”

“…The alteration of MAO-B activity in platelets was reported particularly during migraine attacks, and was more accentuated in women with migraine related to menstrual cycle; this change is not specific for migraine as it was also found in patients with tension-type headache [16].…”

“…In addition to the menstrual cycle hormones, endogenous opioids, serotonin and adrenergic central system of many pure menstrual migraine or menstrually related migraine sufferers may be altered in the premenstrual syndrome 3,27 . Therefore, studies are needed to investigate the pattern of these hormones during the lactation period and their association with the reduction of migraine recurrence during postpartum.…”
