Flutamide is a steroid used to treat androgen-dependent disorders and as antiepileptic, but it induces a number of non-desirable side effects. This work was aimed at assaying the effect of flutamide and two novel synthetic steroids on the levels of GABA, glutamine and oxidative stress markers. Male Wistar rats (weight 180 g) received a single diazepam dose (5 mg/kg) 30 min prior to sacrifice (group A). Group B, flutamide; group C, 16β-methyl-17α-benzoyloxypregnen-4-en-3,20-dione; group D, estrone-3-hemisuccinate; group E, testosterone; group F, progesterone; all administered intraperitoneally at 10 mg/kg, daily for 3 days. Brain and prostate were obtained to assess lipid peroxidation (TBARS), Na(+) , K(+) ATPase activity, reduced glutathione (GSH), γ-amino butiric acid (GABA), glutamine and serotonin (5-HT) concentrations through spectrophotometry, fluorescence and HPLC. GABA levels increased and glutamine decreased in group A (P < 0.05). Total ATPase activity increased in group F and TBARS decreased in group B (P < 0.05). GSH decreased in A, B and C groups. 5-HT increased in group A and the prostate weight was increased in group E. The conclusion is that 16β-methyl-17α-benzoyloxypregnen-4-en-3,20-dione may be considered novel and promising to treat androgen-dependent diseases and epilepsy, since it showed an antioxidant effect and seemed to impair the GABAergic and serotonergic metabolism.

ПАТОГЕНЕТИЧЕСКИЕ АСПЕКТЫ ЭРЕКТИЛЬНОЙ ДИСФУНКЦИИУО ВведениеЭректильная дисфункция (ЭД) определя-ется, как постоянная неспособность достигать Цель. Обобщить и систематизировать имеющиeся в литературе данные о патогенетических меха-низмах развития эректильной дисфункции, что является основой выбора методов лечения и рекоменда-ций для пациентов, имеющих данную проблему.Материал и методы. Для подготовки обзора использовалась база данных PubMed с глубиной поис-ка от 2004 г. по 2014 г. и анализом основных видов публикаций (оригинальные статьи, рандомизирован-ные исследования, обзоры, мета-анализы, рекомендации, консенсусы).Результаты. Проведенные ранее исследования установили высокую распространенность метабо-лического синдрома (МС) и сахарного диабета при эректильной дисфункции (ЭД), а также сходство причин возникновения ЭД и сердечно-сосудистых заболеваний (ССЗ). МС представляет собой комплекс факторов риска, которые потенциально опасны в отношении развития эндотелиальной дисфункции, приводящей к ССЗ и ЭД. Это позволяет предполагать единое объяснение для связи между ЭД, MС и дисфункцией эндотелия. Адипоциты висцеральной ЖТ функционируют как эндокринные клетки, про-дуцируя и секретируя адипокины, а также целый ряд биохимических модуляторов и провоспалительных факторов, влияющих на сосудистое воспаление, что может способствовать развитию ЭД.Лечение ЭД у пациентов с МС должно быть комплексным, и кроме специфической терапии долж-но включать своевременную диагностику и адекватное и активное лечения любых факторов риска, ко-торые могут быть выявлены.Заключение. В патогенезе эректильной дисфункции cocудистого генеза ключевую роль играет со-четание факторов риска, приводящих к патологическим изменениям функции эндотелия сосудов и на-рушению пенильного кровотока. Помимо уже установленных и общеизвестных факторов немаловажное значение может иметь негативное влияние перивисцеральной жировой ткани, что требует дальнейшего изучения. Many signs of aging, such as sexual dysfunction, visceral obesity, impaired bone and muscle strength, bear a close resemblance to features of hypogonadism in younger men. The statistical decline of serum testosterone in aging men is solidly documented. It has been presumed that the above features of aging are related to the concurrent decline of androgens, and that correction of the lower-than-normal circulating levels of testosterone will lead to improvement of symptoms of aging. But in essence, the pivotal question whether the age-related decline of testosterone must be viewed as hypogonadism, in the best case reversed by testosterone treatment, has not been definitively resolved. Studies in elderly men with lower-than-normal testosterone report improvement of features of the metabolic syndrome, bone mineral density, of mood and of sexual functioning. But as yet there is no definitive proof of the beneficial effects of restoring testosterone levels to normal in elderly men on clinical parameters. Few of these studies meet as yet rigorous standards of scientific enquiry: double-blind, placebo-controlled design of the study. The above applies also to the assessment of safety of testosterone administration to elderly men. There is so far no convincing evidence that testosterone is a main factor in the development of prostate cancer in elderly men and guidelines for monitoring the development of prostate disease have been developed. It is of note that there are presently no long-term safety data with regard to the prostate. Polycythemia is another potential complication of testosterone treatment. It is dose dependent and can be managed with dose adjustment.
