Metabolic Restructuring in the Liver under Conditions of Endogenous Intoxication

Abstract: Significant metabolic alterations in the liver were observed in dogs with modeled acute peritonitis. These changes significantly impaired detoxification function of the liver, which was seen from the increase in the titer of toxic products in the early post-surgery period not only in the lymph, but also in the blood plasma. The key pathogenic mechanism leading to acute liver failure is destabilization of cell membrane resulting from LPO, phospholipase activity, and tissue hypoxia. Activation of LPO and increas… Show more

“…Hydrogen peroxide (H 2 O 2 ) is the end product of the SOD reaction can react in vivo to form the hydroxyl free radical, but H 2 O 2 can only be detoxified by the selenoenzyme, glutathione peroxidase (GP), but not by catalase as the Michaelis constant, which is an inverse measure of enzyme affinity, Km (affinity) of GP, for H 2 O 2 is orders of magnitude lower than for catalase [30][31][32]. Data from the scientific literature on the severity of changes not only in biochemical parameters but also in histomorphological changes of hepatocytes in endotoxicosis of various origins were reported [33,34]. Some authors noted that under such conditions, disruption of superoxide dismutase synthesis may be due to reduced production of transport proteins, in particular ceruloplasmin, but these data certainly require a more detailed study [35].…”
