Mesenchymal Stem Cells Improve Wound Healing In Vivo via Early Activation of Matrix Metalloproteinase-9 and Vascular Endothelial Growth Factor

Abstract: We investigated the effects of mesenchymal stem cells (MSCs) on wound healing using a three-dimensional (3D) collagen gel scaffold. Three circular full-thickness skin defects were created on the back of Sprague-Dawley rats. One site was covered with a 3D collagen gel containing 2 × 106 MSCs (MSCs+/3D collagen+). Another site was replaced with a 3D collagen gel without MSCs and the third site was left empty. The wound size was significantly reduced in the MSCs+/3D collagen+ sites. MSCs+/3D collagen+ sites exhib… Show more

“…In diabetic animal models, systemic or local injections, for example, of both MSCs promote angiogenesis and migration, and they have the ability to induce regeneration rather than fi brosis in the wound microenvironment. [94][95][96] . In human diabetic foot ulcers, implantation of autologous BM-MSCs signifi cantly improves clinical parameters such as ulcer size and pain-free walking distance [ 97 ] .…”

“…The characteristic morphology and the molecular level by flow cytometry of MSC cultivated in this study was the same described by other studies for identifying murine MSC (Badillo et al 2007, Falanga et al 2007, Inoue et al 2008, Kwon et al 2008, Mehta et al 2010, Jiang et al 2011, Kim et al 2011.…”

“…This reduction in the fluorescence signal also correlates well with the decrease of the degradation kinetics of the VEGF-loaded hydrogels. The delay in degradation could be attributed to the fact that the biological processes affecting implant resorption in the host are also influenced by the VEGF, including cell-biomaterial interactions and paracrine signaling associated with the implant bioactivity (28,37,40,41). We speculate that the proangiogenic environment induced by the released VEGF at the acute stage of the immune response to the implants (i.e., first week postimplantation) created an environment that would be less accessible to inflammatory cell invasion.…”
