Mesenchymal Stem Cells Augment the Adaptive Response to Eccentric Exercise

Abstract: The results from this study demonstrate that mMSCs contribute to beneficial changes in satellite cell expansion and growth in α7Tg muscle after eccentric exercise. Thus, MSCs that naturally accumulate in the muscle after eccentric contractions may enhance the adaptive response to exercise.

“…To quantify myofiber CSA, dystrophin-Alexa Fluor 633 images were imported into Adobe Photoshop (CS5 Extended; Adobe, San Jose, CA, USA), and an average of 400 fibers were circled using the magnetic lasso tool, which decreases subjectivity and interassessment error (26). To determine MHC myofiber CSA, ;100-150 myofibers per type were circled on average as described above.…”

“…Overall, these findings suggest that manipulation of SCs may not be the optimal approach to improve recovery postdisuse. Alternative stem and stromal cells, including mesenchymal stem/stromal cells and pericytes, exist in skeletal muscle that similarly demonstrate capacity for repair following injury, yet the ability of these cells to induce growth and recover mass following disuse remains unexplored (23)(24)(25)(26).…”

“…137,146 Interestingly, transplantation of young (5 week) musclederived mMSCs (1 Â 10 4 ) into recipient muscle 1 h following an acute bout of exercise (protocol described above) significantly increased satellite cell number and new fiber synthesis 141 In addition, transplantation increased myonuclear accumulation, myofiber growth, and strength following repeated bouts of eccentric exercise (3Â/week, 4 weeks) in adult mice. 147 In both studies, mMSCs did not express Pax7, form new fibers, or fuse with existing fibers. The fact that transplanted cells localized to areas of CLN + fibers and vessels and contributed to vessel remodeling following an acute bout of exercise 148 suggests the ability of mMSCs to repair multiple tissues via release of local factors.…”
