Meristem-specific gene expression directed by the promoter of the S-phase-specific gene, cyc07, in transgenic Arabidopsis

Abstract: A genomic clone for the cyc07 gene, which is expressed specifically at the S phase during the cell cycle in synchronous cultures of periwinkle (Catharanthus roseus) cells, was isolated. Determination of the nucleotide sequence of the clone revealed that the cyc07 gene consists of seven exons separated by six introns. Genomic Southern analysis indicated that the cyc07 gene is present as a single copy per haploid genome in periwinkle. Expression of related genes was detected in a wide range of other plants. Tran… Show more

“…Whereas OCEs and Octs are highly conserved among histone genes, they are rarely found in other genes, including those which are (expected to be) expressed at the G1/S boundary and S phase, such as the PCNA gene (Kosugi et al . 1991) and the cyc07 gene (Ito et al . 1994).…”

Recently, we found a type I-like element in the promoters of a gene encoding the non-histone chromatin protein HMG-I/Y (Gupta et al, 1998) and a related gene (G. Morisawa et al, unpublished data).…”

“…Transcriptional start sites of CYS and CYM were determined by primer extension analysis using oligonucleotide primers corresponding to the nucleotide positions 23-50 of the CYS cDNA and 20-48 of the CYMcDNA. Primer extension analysis was performed as described previously (Ito et al, 1994).…”

“…Cyc07 genes from Catharanthus roseus [25] and S. cerevisiae [26] have been reported to be expressed in S phase and their products have been proposed to play a role in cell proliferation. C. roseus cyc07 is expressed mainly in the meristematic regions of transgenic Arabidopsis [31]. In addition, cyc07 expression is regulated by environmental stress, including high osmotic pressure, salinity, low temperature and submergence in Oryza sativa [27].…”
