Heavy metals, including mercury (Hg) and cadmium (Cd), occur naturally or anthropogenically and are considered toxic to the environment and human health. However, studies on heavy metal contamination focus on locations close to industrialized settlements, while isolated environments with little human activity are often ignored due to perceived low risk. This study reports heavy metal exposure in Juan Fernandez fur seals (JFFS), a marine mammal endemic to an isolated and relatively pristine archipelago off the coast of Chile. We found exceptionally high concentrations of Cd and Hg in JFFS faeces. Indeed, they are among the highest reported for any mammalian species. Following analysis of their prey, we concluded that diet is the most likely source of Cd contamination in JFFS. Furthermore, Cd appears to be absorbed and incorporated into JFFS bones. However, it was not associated with mineral changes observed in other species, suggesting Cd tolerance/adaptations in JFFS bones. The high levels of silicon found in JFFS bones may counteract the effects of Cd. These findings are relevant to biomedical research, food security and the treatment of heavy metal contamination. It also contributes to understanding the ecological role of JFFS and highlights the need for surveillance of apparently pristine environments.

Pengujian Metil Merkuri (Me-Hg) dalam Matrik Biomarker menggunakan Kromatografi Gas Electron Capture Detector (GC-ECD). Merkuri (Hg) merupakan salah satu logam berat yang sangat berbahaya bagi kesehatan dan lingkungan hidup karena bersifat toksik, persisten, bioakumulasi dan dapat berpindah dalam jarak jauh di atmosfer. Hg dapat berupa senyawa anorganik dan organik. Salah satu senyawa merkuri organik berbahaya adalah metilmerkuri (Me-Hg). Me-Hg terbentuk dari merkuri anorganik oleh aktivitas organisme anaerob dalam sistem perairan. Pengujian Me-Hg dalam matriks biomarker dilakukan pada sampel rambut manusia, sebagai bagian dari kajian pencemaran lingkungan. Manusia terkontaminasi Hg dari berbagai sumber, namun apabila melalui jalur makanan biasanya pengujian Me-Hg ditentukan melalui pengukuran sampel rambut. Laboratorium Pusat Standardisasi Instrumen dan Kualitas Lingkungan Hidup (PSIKLH) melakukan validasi pengujian Me-Hg dalam sampel biomarker berupa rambut dengan mengadopsi metode yang dilakukan oleh Laboratorium IDEA Consultant Inc. Jepang. Kegiatan dilakukan pada bulan Mei -Desember 2022. Terdapat penggantian pelarut dari toluena menjadi dichloromethane (DCM) dan juga terdapat perlakuan awal terhadap kolom dengan penambahan Methanolic-hydrobromic acid (MeOH-HBr) 0,3 mM. Validasi dilaksanakan dengan memperhatikan aspek penentuan kurva kalibrasi, linieritas, limit deteksi metode dan limit kuantifikasi, akurasi, presisi, dan ketahanan metode. Pengujian dilakukan dengan menggunakan CRM NIMD 01 yang memiliki kisaran konsentrasi 0,563 ± 0,0705 mg/kg. Hasil validasi laboratorium PSIKLH menunjukkan limit deteksi sebesar 0,08 mg/kg, limit kuantifikasi sebesar 0,3 mg/kg, dengan nilai akurasi dan presisi memenuhi keberterimaan yang dipersyaratkan.

Mercury (Hg) is a chemical element that poses risks to human health due to its high toxicity and environmental persistence. We determined the total Hg (THg) and methyl Hg (MeHg) concentrations in hair samples from residents of the Demarcação District (Porto Velho, Rondônia) in the Brazilian Amazon, as well as in water and fish samples, to evaluate factors influencing human exposure. The average THg concentration in human hair was 7.86 ± 6.78 mg kg−1 and it was significantly higher in men, with an increasing trend related to age. There was no significant difference between female age groups. Human exposure to Hg through water was negligible compared to fish consumption. The average weekly intake estimates in the community varied between 1.54 and 4.62 μg kg−1, substantially higher than the recommended limit. The fish species with the highest amounts safe for daily consumption were herbivores and detritivores. Our results contribute to an understanding of how exposure to Hg affects the health of riverside populations and provide insights for new research to develop methods to mitigate such exposure and thus improve the quality of life of Amazonian people.
