“…This hypothesis is supported by two of the five top terms in the enrichment analyses, “above average bilayer thickness” and “high bilayer thickness.” Out of the 147 dysregulated lipids, 60.5% (89/147) were phospholipids, which are primarily located within cellular membranes (plasma membrane and organelle membranes). Although the lipid composition of each cellular membrane has a different abundance in lipid species, 129 we did not explore which cellular membrane (plasma membrane, endoplasmic reticulum, Golgi, etc.) may be most affected by TBT exposure.…”

“…Last, the term “negative intrinsic curvature” indicates that TBT may induce a concave membrane structure, which is driven by higher abundance of phosphatidylethanolamines and phosphatidic acid. 129 We hypothesize that the combined reduction in phosphatidylcholines, phosphatidylserines (drivers of no membrane curvature), and phosphatidylethanolamines results in membrane curvature changes. The direct impact of curvature changes upon TBT exposure on cell function, however, remains to be determined.…”

“…PG is transported to the inner mitochondrial membrane and acts as a precursor for cardiolipin biosynthesis, a lipid found exclusively within mitochondria. PG is a relatively rare lipid species in mammalian membranes and is more abundant in bacterial membranes (Sim et al, 2023). Vancomycin may therefore localise to mitochondria in mammalian cells via the increased abundance of PG used for cardiolipin synthesis, however we also saw significant labelling of other membrane structures including the plasma membrane and nucleus.…”

“…The composition of membranes differs between intracellular organelles (Sim et al, 2023), and these differences may affect vancomycin binding in different cell types, but this may also be influenced by additional factors such as shape, charge and presence of transmembrane proteins. The function and integrity of mitochondria depends on the composition of lipids that make up their membranes.…”
