Melatonin Modified Release Formulations Designed for Sleep Disorders

Abstract: Melatonin (N-acetyl-5-methoxytryptamine, MLT), a hormone synthesized by the pineal gland and released at night, has a regulatory role on sleep in vertebrates, including humans. It has been shown to have a hypnotic action in animals and humans and it has been used as an agent for restoring circadian rhythms, disturbed by jet-lag, shift-work or aging. The physiological actions of melatonin in regulating seasonal and circadian rhythms are mediated through a family of specific, high affinity G protein-coupled memb… Show more

“…To address this, nanotech ophthalmic systems could be considered to traverse the ocular surface epithelial barrier and reach tissues within the eye globe. Several nanocarriers have been described for ocular drug delivery to different eye segments [ 22 ], and some of them have shown promising results in improving ocular bioavailability, reducing the frequency of administration, and minimizing toxic effects on healthy tissues ( Table 1 ).…”

“…A singlewalled carbon nanotube (SWCN) improved bioabsorption of zaleplon (sedative-hypnotic) via nasal delivery, by increasing carbon nanotube-mediated bloodbrain crossing ability [417], leading to prolonged neuroprotective effect. Melatonin is another suitable therapeutic candidate for insomnia which could be transdermally delivered due to its low molecular weight [415,[418][419][420]. A microneedle patch synthesized from proline, melatonin and silk fibroin has been found effective in maintaining drug concentration in blood and demonstrates physiological resemblance in drug release in Sprague-Dawley rat models of insomnia [416].…”

“…Therefore, the modified release of MLT from oral solid dosage forms that alter the time and/or the rate of MLT release may provide an alternative for MLT delivery and be useful in the treatment of circadian rhythmic disorders, like insomnia, jet lag, seasonal affective disease, shift work syndrome, etc. [6]. The recent advantages of MLT modified-release per oral solid formulations are reviewed below and summarized in Table 1.…”
