Melanin Produced by Bordetella parapertussis Confers a Survival Advantage to the Bacterium during Host Infection

Abstract: In addition to the Gram-negative bacterium Bordetella pertussis , the etiological agent of pertussis, Bordetella parapertussis also causes respiratory infection in humans, with a mild pertussis-like disease. These bacteria are genetically closely related and share many virulence factors, including adhesins and toxins.

“…Transposon-based integration of the Km r -P tac -GFP fragment into the B. bronchiseptica genome was confirmed by colony PCR using the primers BS333-F and BS334-R. The integration site of Km r -P tac -GFP was determined as previously described ( 44 ). Briefly, genomic DNA (gDNA) of a single mutant was isolated using the DNeasy Blood and Tissue kit (Qiagen) and digested with Sau3AI (TaKaRa).…”

“…The Bvg + phase is generally considered to represent the virulent phenotype, while Bvg − phase represents the avirulent phenotype. The Bvg − -locked mutant of B. parapertussis , which constitutively exhibits the Bvg − phenotype ( 10 , 11 ), did not increase Bdk-induced cAMP production ( Fig. 2 A ).…”

“…A Himar1‐based transposon vector, pMariG, 11 was introduced into B. bronchiseptica RB50 via biparental conjugation through E.coli S17‐1 λpir. After a 6‐h incubation, the bacteria in the mixture were collected and cultured on BG plates supplemented with Gm for 48 hr at 37°C.…”
