Six polysaccharide-producing bacteria, isolated from soil samples and identified as Pseudomonas and Arthrobacter (Strains 3B, 4B, 7B, 21B, 18E and 21D), were tested for the yield of polysaccharides produced during growth in two culture media: one containing glucose and the other sucrose (1, 2, 3, 4 and 5%). The yield was quantified measuring the viscosity of the broth, using the Poiscuille equation. The effect of temperature and pH of the culture media was investigated. The largest polysaccharide yield was obtained when the concentration of the carbon source was lower than 2%. Glucose and sucrose stimulated the polysaccharide production in a similar way. When the initial pH of the fermentation broth was increased from 5.0 up to 7.0, there was an increase in polysaccharide production. However, higher values (pH=8.0) caused a decrease in polysaccharide production. With regard to temperature, 30ºC was shown to be optimum, since higher or lower temperatures had a negative effect on saccharide production.

Solid state fermentations were carried out using a strain of Rhizopus formosa MUCL 28422 for its ability to grow on raw cassava. Optimal substrate composition, as estimated by surface response design experiments, was 10% each of cassava bagasse and soybean¯our and 80% cassava¯our. Optimal fermentation conditions were: temperature 32°C, initial pH and moisture of the substrate 6.5 and 64%, respectively, and inoculum size 10 6 -spores g À1 dry matter. These conditions were employed for studying the kinetics of the biotransformation of cassava¯our using Oxygen Uptake Rate (OUR) and Carbon dioxide Evolution Rate (CER) during the process. The respiratory quotient was $1, corresponding to an aerobic process in the ®rst 24±26 h. Sporulation started after 26 h of fermentation when the respiratory quotient showed a trend to increase up to $6. Biomass estimation, based on oxygen consumption (Y x/o ) gave a value of 0.531 g biomass g À1 O 2 consumed and the maintenance coef®cient (m) was of the order of 0.068 g O 2 consumed g À1 biomass h À1. The corresponding value for the speci®c growth rate (m) was 0.16 h À1.

The Plackett-Burman screening method was utilized as a tool to evaluate the importance of the selected six factors, including methanol, ethanol, propanol, butanol, pH and time, which are relevant to the extraction of the antimicrobial substance produced by Bacillus sp. fmbJ. The main factors that affected the extraction of the antimicrobial substance were determined as methanol (P < 0.0001), ethanol (P < 0.0001), pH (P ¼ 0.0032), and time (P < 0.0001) by using the JMP software. Within the test ranges, methanol, ethanol, and time showed a significant positive relativity to the total extracted amounts respectively; while pH had a significant negative effect. The maximum prediction profile indicated that the total extracted amounts for the antimicrobial substance would reach 50.21 mg/100 ml with 99.64% probability.
