Mechanism of refractory ceramic fiber- and rock wool-induced cytotoxicity in alveolar macrophages

Abstract: These results suggest that RF2 and RW1 can induce cytotoxicity and ATP depletion in the AM through the polyinosinic acid-inhibitable scavenger receptor. ATP depletion was the important factor in MMVF cytotoxicity, especially by RW1.

“…The changes in the levels of transaminases are the indicators of impaired liver function state [45,46] . Kim et al [47] have suggested that the significant increase in the activities of hepatic marker enzymes such as AST, ALT and ALP manifested by CsA induced hepatocellular damage. Administration of propolis in rats significantly decreased the activities of AST and ALT (P<0.01) ( Table 4), suggesting that they offer protection by preserving the structural integrity of the hepatocellular membrane against CsA [48] .…”

“…Slag wool proved the most toxic, while RCF 1 induced the lowest LDH release [92]. However, another refractory ceramic (RCF 2) was found to be cytotoxic, as shown by LDH release as well as by ATP depletion in rat AMs [88]. This apparently contradictory result was due to fiber composition, as RCF 1 was made from kaolin and RCF 2 from zirconium [92], indicating a role of fiber components in cytotoxicity.…”

“…As for apoptosis, this may be fiber length, as fiber D sample contained the higher proportion of fibers over 20 µm long. Moreover, this effect increased with the time of cell exposure to fibers [34,88]. In another study, long and thick glass fibers designed for building insulation displayed a weak toxic effect [89].…”

“…The measurement of total lactate dehydrogenase can be useful when only a specific organ, such as the liver, is known to be involved. LDH is a sensitive intracellular enzyme, which can be used as an indication in liver cell damage (Kim et al 2001). Therefore, the extent of cellular damage was measured in terms of release of leakage enzymes including AST, ALT and LDH.…”
