SummaryThis study was planned to determine the effects of propolis in rats applied Cyclosporine A (CsA). In this study, 24 male Sprague-Dawley rats were used. Rats were randomly divided into 4 groups including control and 3 treatment groups. Group 1 (Control) were no supplement; CsA (group 2) were given as s.c. 15 mg/kg body weight (BW) every day; Propolis (group 3) were given by gavage 100 mg/kg BW every day; CsA+Propolis (group 4) were given as s.c. 15 mg/kg BW of CsA and by gavage 100 mg/kg BW of propolis every day. The feed intake were significantly higher (P<0.01) in Control and Propolis groups than CsA and CsA+Propolis groups within time period of 21 days. Further, body weight was significantly lower (P<0.01) in groups administrated with CsA (Group 2 and 4) than the other groups. Cortisol, AST, ALT and urea levels in serum of Control, Propolis and CsA+Propolis groups were found significantly lower (P<0.01) than those of CsA group. Malondialdehyde levels in kidney and liver tissues were significantly higher (P<0.01) than in the CsA groups compared to other groups. The catalase and reduced glutathione activities in kidney tissue of CsA+Propolis group were significantly higher (P<0.01) than those of CsA group. The present study demonstrated that propolis provided amelioration in terms of hepatotoxicity and nephrotoxicity consisting rats applied to CsA.

The ultimate goal of toxicologic investigation of synthetic vitreous fibers (SVFs) is to provide essential input for the assessment of human risk to their exposure. Toxicity of mineral fibers is usually evaluated by testing biopersistence in rodent model. However, a cellular model would be much appreciated in order to reduce, refine, and replace animal models. Pulmonary disorders triggered by inhalation of occupational or environmental mineral particulates can be the endpoints of a chronic inflammatory process in which alveolar macrophages (AMs) play a crucial role. Depending on the type of SVF involved, phagocytosis of fiber leads to activation of macrophages, resulting in release of fiber components and potent mediators, such as reactive oxygen or nitrogen species and cytokines. As a matter of fact, macrophages should be the cells of choice since SVF toxicity is the consequence of fibers and alveolar macrophages interaction. Today, monocytes and macrophages culture are firmly established as a paradigm in toxicology when several endpoints are assayed in macrophages: (1) fiber durability, (2) fiber surface changes, (3) oxidative stress and genotoxicity in macrophage, and (4) macrophage cell viability and apoptosis. This article is a review of up-to-date knowledge of in vitro studies involving macrophages, and assesses endpoints of macrophage toxicity with an emphasis on (1) dissolution, (2) scanning electron microscopy analysis, (3) cytotoxicity, and (4) gene expression.

Objective: The objective of this study is to better understand the hepatoprotective effect of GTs and to enhance their use in food supplement pharmaceutical and medical industries. Materials and methods: HepG2 cells were pretreated in the presence or absence of GTs (50, 100 and 200 mg/ml) for 4 h, then exposed with 60 mmol/L of t-BHP for an additional 4 h. The cell viability was evaluated by MTT method. ALT, AST and LDH production in culture medium and intracellular MDA, GSH and SOD levels were determined. Moreover, the total triterpenoid content and chemical constituents in GTs were detected by ultraviolet spectrophotometry and HPLC/Q-TOF-MS, respectively. Results: GTs (50, 100 and 200 mg/ml) significantly increased the relative cell viability by 4.66, 7.78 and 13.46%, respectively, and reduced the level of ALT by 11.44%, 33.41% and 51.24%, AST by 10.05%, 15.63% and 33.64%, and LDH by 16.03%, 23.4% and 24.07% in culture medium, respectively. GTs could also remarkably decrease the level of MDA and increase the content of GSH and SOD in HepG2 cells. Furthermore, the total triterpenoid content in GTs was 438 mg GAAEs/g GTs. And 16 triterpenoids in GTs were identified or tentatively characterised. Discussion and conclusion: Our results showed that GTs had potent cytoprotective effect against oxidative damage induced by t-BHP in HepG2 cells, thus suggesting their potential use as liver protectant.ARTICLE HISTORY

Abstract: These results suggest that RF2 and RW1 can induce cytotoxicity and ATP depletion in the AM through the polyinosinic acid-inhibitable scavenger receptor. ATP depletion was the important factor in MMVF cytotoxicity, especially by RW1.
