Mechanism of NF-κB signaling pathway and autophagy in the regulation of osteoblast differentiation

Abstract: The osteoblast itself had a strong function of autophagy. The appropriate concentration of SN50 in blocking the NF-κB pathway of the osteoblast was associated with the obvious inhibition of autophagy. However, the relationship between NF-κB signaling pathway and autophagy in the process of tooth eruption requires further study.

“…We previously reported that osteoblasts themselves robustly induce autophagy, and the expression of autophagyrelated proteins was altered significantly by the action of NF-kappa B inhibitor SN50. The results suggested that the autophagy function of osteoblasts during tooth eruption plays an important role through the NF-kappa B signaling pathway (Qin et al, 2016). As an important regulator of osteoblasts, it is important to determine how POSTN relates to the autophagy function of osteoblasts and how it regulates the dynamic balance between osteoblasts and osteoclasts.…”

“…Previous studies have shown that cortical bone density and bone strength have decreased in POSTN-knockout mice, but the specific mechanism is not yet clear (Bonnet et al, 2013). Our previous studies have shown that osteoblasts themselves robustly induce autophagy (Qin et al, 2016). However, the effect of autophagy on osteoblasts during tooth eruption and the association between this effect and tooth eruption are unknown.…”

“…SN50, a blocking agent of the NF-κB pathway, exerts a significant effect on autophagy in osteoblasts only when administered at a certain concentration (25 mg/mL). At this dose, it significantly inhibited autophagic flux in osteoblasts by blocking the NF-κB pathway (Qin et al, 2016). Tnf-α promoted osteoblast autophagy and reduced osteoblast apoptosis in avascular necrotic tissue in the femoral head by activating the P38 MAPK/NF-κB signaling pathway (Zheng et al, 2020).…”

“…Osteoblasts’ autophagy-related function changes significantly when the NF-κB signaling pathway is disabled, demonstrating a possible connection between the two mechanisms 220 . However, according to some studies, NF-κB appears to play two roles in autophagy.…”
