Mechanism of Inhibition of P-Glycoprotein Mediated Efflux by Vitamin E TPGS:  Influence on ATPase Activity and Membrane Fluidity

Abstract: Efflux pump (e.g., P-gp, MRP1, and BCRP) inhibition has been recognized as a strategy to overcome multi-drug resistance and improve drug bioavailability. Besides small-molecule inhibitors, surfactants such as Tween 80, Cremophor EL, several Pluronics, and Vitamin E TPGS (TPGS 1000) are known to modulate efflux pump activity. Competitive inhibition of substrate binding, alteration of membrane fluidity, and inhibition of efflux pump ATPase have been proposed as possible mechanisms. Focusing on TPGS 1000, the aim… Show more

“…Dans le même esprit, il nous a semblé que le recours à un système d'administration contenant du poly (éthylène glycol) pouvait être intéressant. Mises à part ses propriétés solubilisantes, cet excipient possède en effet une l'activité inhibitrice de la P-gp et du cytochrome P450 [31,32]. De plus, il est connu que la pégylation des nanoparticules leur confère des propriétés lubrifiantes et muco-adhésives.…”

“…27 Efflux proteins show a strong efflux effect in low drug concentrations, and become saturated in high drug concentrations. When the Lac-NCTD concentration increased from 100 µg/mL to 400 µg/mL, the ratios of Papp BL-AP to Papp AP-BL decreased from 1.91 to 1.37, respectively.…”

“…Then research was preceded by Woodcock et al (1992) which then conclude some other nonionic surfactants, e.g. Tweens, Spans, Cremophors (EL and RH40), Pluronics and vitamin E TPGS having P-gp inhibition Ho et al (2007) action (Collnot et al, 2007). Cremophor EL was used as a component of marketable formulations of paclitaxel (Taxol), however this formulation was found to be toxic.…”
