Mechanism of Endothelial Nitric Oxide-Dependent Vasorelaxation Induced by Wine Polyphenols in Rat Thoracic Aorta

Abstract: The mechanisms by which red wine polyphenolic compounds (RWPCs) induced endothelium-dependent relaxation were investigated in rat thoracic aorta rings with endothelium. RWPCs produced relaxation that was prevented by the nitric oxide (NO) synthase inhibitor, N(omega)-nitro-L-arginine-methyl-ester. This relaxation was abolished in the absence of extracellular calcium in the medium or in the presence of the Ca2+ entry blocker, La3+, but it was not affected by the nonselective K+ channels blocker, tetrabutylammon… Show more

“…This result is well consistent with report that PCRW produced the endothelium-NO-dependent relaxation through an extracellular Ca 2+ -dependent mechanism (Andriambeloson et al, 1999). Amongst the different classes of polyphenolic compounds present in PCRW, anthocyanins and oligomeric condensed tannins had the same pharmacological profile as PCRW (Andriambeloson et al, 1998).…”

“…Ethanol, at the same concentration as contained in red and white wines, does not cause vasorelaxation (31). RW or extracts obtained from it caused endothelium-dependent, NO-mediated vasorelaxation of rat (32) or rabbit (33) aorta preconstricted with norepinephrine. Also, in human coronary arteries and rat aortic rings in vitro, short-term incubation with RW induced FMD and increased vascular cGMP content (31).…”

“…This role of Ca 2 + is supported by the relaxation of rat aorta (64) and the increase in Ca 2 + concentration in bovine aortic endothelial cells (65) caused by a polyphenolic extract from alcohol-free red wine and by delphinidin, which are both Ca 2 + dependent effects. Furthermore, wine polyphenols are unable to increase Ca 2 + concentration in smooth muscle cells, indicating the selectivity of this effect (65) .…”
