Measurement of Refractive State and Deprivation Myopia in Two Strains of Mice

Abstract: The infrared refraction technique is sufficiently sensitive to resolve equivalent changes in axial length of only +/- 10 microm in alert mice. Prolonged occlusion produces a significant myopic shift in B6 mice, but not in D2 mice. Even among isogenic B6 mice, the response is variable for reasons that presumably trace back to subtle developmental, environmental, and technical factors.

“…33,37 Briefly, each unanesthetized mouse was placed on a platform that was slowly adjusted until a clear first Purkinje image occurred in the center of the pupil, indicating an on-axis measurement. The data were then automatically recorded by the program designed by Schaeffel 33 and reported as the mean of at least three measurements, each of which was averaged from 10 individual values to create that measurement.…”

“…The refractive state was measured by an automated infrared photorefractor in a dark environment as described previously. 33,34 Refractive error values were recorded with a custom software designed by Frank Schaeffel. For each eye, an average of 50 to 100 measurements were used for analysis.…”

“…Form-deprivation myopia was induced by the placement of a translucent occluder onto a randomly selected eye for 4 weeks, as described previously by Schaeffel et al 36 D1R antagonist, SCH39166 solution (0.04 lg/lL; Tocris bioscience, Glasgow, UK) was freshly prepared by dissolving it in dimethyl sulfoxide (DMSO; 0.11 lg/lL; Sigma, Buchs, Switzerland). SCH39166 (0.4 lg/g body weight), or DMSO (1.1 lg/g bodyweight) were intraperitoneally injected daily without anesthesia to drug-injection groups.…”
