Measurement of leptin in dried blood spot samples

Abstract: Leptin is important in a wide range of physiological processes, but logistical constraints associated with venipuncture blood collection have limited research on leptin in diverse, community-based settings. The aim of this short report is to present and validate an enzyme immunoassay method for quantifying leptin in samples of capillary whole blood collected from a simple finger prick and dried on filter paper. The method was evaluated through analysis of precision, reliability, stability, and comparisons with… Show more

“…We did, however, find that DBS values were consistently lower than serum or plasma values. Similar differences in serum or plasma versus DBS levels have been reported previously for CRP and other analytes ((McDade and Shell-Duncan, 2002; McDade et al, 2004; Miller et al, 2006; Baingana et al, 2008) and others). These differences might stem from inaccuracy in the estimate of sample volume contained in DBS punches (Mei et al, 2001), or might indicate incomplete elution of the CRP from the paper.…”

“…Proteins in blood spots are beyond detection in a matter of days [4], but nucleic acids show evidence of long-term stability. The published literature shows extensive examples of recovery and analysis of nucleic acids from dried blood spots.…”

“…This is an expanding area of research as assays for more analytes are currently being developed 1 . Among the assays already validated and being widely used are cholesterol 2 ,CRP 10,14,15 , IgE16, EBV 9,15 , leptin 17 , and cyotkines 18 . There are a number of laboratories specializing in the assay of DBS, including the University of Washington Department of Laboratory Medicine (UW Lab Med) Biomarker Laboratory (Director: Mark Wener).…”
