Intact follicles of the bambooleaf wrasse were incubated with different radioactively labeled steroid precursors during final oocyte maturation (FOM), and the maturation‐inducing hormone (MIH) of this species was examined to compare the activities of the steroid metabolites produced on germinal vesicle breakdown (GVBD) in vitro. Of the metabolites obtained by thin layer chromatography, two steroids, 17,20β‐dihydroxy‐4‐pregnen‐3‐one (17,20β‐P) and 17,20β,21‐trihydroxy‐4‐pregnen‐3‐one (20β‐S), both had the greatest effect on GVBD in vitro. During FOM, a shift in steroidogenic enzymes, a decrease in C17,20‐lyase activity, and an increase in 20β‐HSD activity were found. In addition to this shift, continuous high 21‐hydroxylase activity throughout all the oocyte developmental stages was observed, and, in all likelihood, this 21‐hydroxylase activity enables ovarian follicles to synthesize enough 17,21‐P. Consequently, activated 20β‐hydroxysteroid dehydrogenase converted 17‐P and 17,21‐P to 17,20β‐P and 20β‐S, respectively, during FOM. Thus, the present study not only provides evidence on the physiological role of 17,20β‐P and 20β‐S as MIH in the bambooleaf wrasse, but also details the enzymatic kinetics of 17,20β‐P and 20β‐S biosynthesis.

Teleost fishes are a large and diverse animal group that represent close to 50% of all described vertebrate species. This review consolidates what is known about the claudin (Cldn) family of tight junction (TJ) proteins in teleosts. Cldns are transmembrane proteins of the vertebrate epithelial/endothelial TJ complex that largely determine TJ permeability. Cldns achieve this by expressing barrier or pore forming properties and by exhibiting distinct tissue distribution patterns. So far, ~63 genes encoding for Cldn TJ proteins have been reported in 16 teleost species. Collectively, cldns (or Cldns) are found in a broad array of teleost fish tissues, but select genes exhibit restricted expression patterns. Evidence to date strongly supports the view that Cldns play a vital role in the embryonic development of teleost fishes and in the physiology of tissues and organ systems studied thus far.

Atlantic cod Gadus morhua ovaries were incubated in vitro with tritiated 17-hydroxypregn-4-ene-3,20-dione (17-P) to determine whether 17,20beta-dihydroxypregn-4-en-3-one (17,20beta-P) or 17,20beta, 21-trihydroxypregn-4-en-3-one (17,20beta,21-P), or both, were more likely to be the steroid responsible for inducing oocyte final maturation (i.e. resumption of meiosis). Only 17,20beta,21-P was produced, in addition to 11-deoxycortisol (17,21-P), which is intermediate between 17-P and 17,20beta,21-P. Also, the 5beta-reduced forms of 17-P, 17,21-P and 17,20beta,21-P were all found. Some sulphation of 21-hydroxylated steroids was demonstrated. The ability of female G. morhua to make 17,20beta,21-P but not 17,20beta-P was confirmed by radioimmunoassay of plasma samples from spawning fish. Although small amounts of 17,20beta-P immunoreactivity were detected in a few plasma samples, this was shown, by thin-layer chromatography, to be mostly due to cross-reaction with other unidentified compounds. The evidence strongly suggests that 17,20beta,21-P is more likely than 17,20beta-P to be the maturation-inducing steroid in G. morhua.
