Matrix Metalloproteinase 9 (MMP-9)-dependent Processing of βig-h3 Protein Regulates Cell Migration, Invasion, and Adhesion

Abstract: Background: Cell migration is involved in altering the cell and matrix interface on the cell surface. Results: ␤ig-h3 is cleaved by MMP-9, and its cleavage results in changes in its binding properties, cell adhesion, cell migration, FAK/Src signals, and chemoattractant effects. Conclusion: MMP-9-cleaved ␤ig-h3 modulates tumor cell and macrophage migration. Significance: The MMP-9-mediated ␤ig-h3 processing mechanism is crucial for understanding cell migration.

“…43 MMP9 has previously been identified as a downstream target of Src/FAK signaling. 44 Consistent with this and with the critical role of MMP9 in invasion, teriflunomide may suppress TNBC invasion by blocking MMP9 expression. Several studies linked TNBC to some growth factor receptors, such as EGFR 45 and IGF1R, 46 which were considered as major oncogenic factors and attractive therapeutic targets in the management of TNBC patients.…”

“…MMP-9 production and activity could be induced by LPS, which is a common potent activator of inflammatory responses [22,23,24,25]. In addition, MMP-9 was closely associated with cell migration [4,5]. In the current study, we verified that LPS not only induced the production of MMP-9 mRNA and protein expression, but also induced the potential activity of MMP-9, as well as cell migration.…”

“…Since MMPs, especially MMP-9, are required for cell migration [3,4], we assessed the migration of RAW264.7 cells. LPS at 25-200 ng/mL induced a significant increase in cell migration following 24-h incubation (Fig.…”

“…TGFβ-induced protein (TGFBI, aka βig-h3) is a 68 kDa RGD motif-containing protein that binds to type I, II and IV collagens. This protein is induced by TGFβ and acts to inhibit cell adhesion, cell-ECM interaction and cell migration [49;50]. Various mutations are associated with corneal dystrophy.…”
