Acetylcholine Inhibits LPS-Induced MMP-9 Production and Cell Migration via the a7 nAChR-JAK2/STAT3 Pathway in RAW264.7 Cells

Abstract: Background: Excessive activation of matrix metalloproteinase 9 (MMP-9) has been found in several inflammatory diseases. Previous studies have shown that acetylcholine (ACh) reduced the levels of pro-inflammatory cytokines and decreased tissue damage. Therefore, this study was designed to explore the potential effects and mechanisms of ACh on MMP-9 production and cell migration in response to lipopolysaccharide (LPS) stimulation in RAW264.7 cells. Methods: MMP-9 expression and activity were induced by LPS in RA… Show more

“…This provides a possible mechanistic explanation for the role of LIPUS in HAECs migration through their regulatory function in type I and type III collagens degradation. Furthermore, among different types of MMPs, MMP-1, MMP-2, MMP-9 and MMP-12 are the crucial factors both in sustaining cell basement membrane structure and promoting migration [55, 56]. Our study showed that LIPUS stimulation down-regulated the levels of MMP-1, MMP-2, MMP-9 and MMP-12 under oxidative stress condition.…”

“…Superoxide generation was determined by fluorescent-labelled dihydroethidium (DHE, Molecular Probes) staining as previously described [22,28]. Protein immunofluorescence staining was performed as previously described [21,29]. The primary phosphorylated IKKβ (p-IKKβ, a marker of NF-κB activation), TNF-α and NAD(P)H oxidase subunit gp91 phox antibodies were from Santa Cruz Biotechnology.…”

“…Numerous reports have shown that this rupture depends on the destruction of the extracellular matrix (ECM) by MMPs [2]. MMP-9, one of the most prevalent MMPs produced by monocytes/macrophages, is highly expressed in the vulnerable regions of advanced atherosclerotic plaque, and it has been suggested it is associated with plaque progression and destabilization [3][4][5][6].…”
