Mapping of antigenic sites on the bovine ephemeral fever virus glycoprotein using monoclonal antibodies

Abstract: Monoclonal antibodies (MAbs) were produced against the G, M2 and N proteins of bovine ephemeral fever virus (BEFV) and 29 were selected for further study. Thirteen neutralizing MAbs were assigned to one conformation-independent and at least two conformation-dependent antigenic sites on the G protein by a competitive binding ELISA. The panel of MAbs were tested by neutralization and immunofluorescence with three strains of BEFV and three BEFV-related viruses. The results indicated that BEFV strains from differe… Show more

“…Results showed a 97.4-99.6% homology between the 1983 and 1999 isolates, which led to alterations in 4 amino acids in antigenic sites G1, G3b, and G3c (Table 4). 6,10,25 Results also showed a 96.7-97.3% homology between Taiwanese isolates and the Japanese strains and a 90.3-90.6% homology with the BB7721 strain (Fig. 2).…”

“…Competitive binding assays and the location of neutralization escape mutations indicate that amino acid residues D &( , R #") and Q #'& are closely associated in site G3a, and that residues Q %* and E ##* are closely associated in site G3b. (Cybinski et al, 1990 ;Kongsuwan et al, 1998 ; Fig. 3 A).…”

“…3 A). Indeed, all escape mutations in site G3a could be selected using a single MAb (3D6), and the site G3b mutations were selected using two MAbs (16A6 and 8D3) which compete strongly for epitope binding (Cybinski et al, 1990). To achieve appropriate proximity of these residues, it appears essential that the first linear element of the site (C I to C II ) should be antiparallel to the second element (C VIII to C IX ), and that the third linear element (C X to C XI ) should then be parallel to the first.…”

“…0000-9805 © 1991 SGM including a trypsin-sensitive glycoprotein, an NS protein and two matrix proteins. We have recently described the antigenic structure and properties of the virion glycoprotein which has at least three antigenic sites containing neutralizing and protective epitopes (Cybinski et al, 1990). In this paper, we have re-examined the virion proteins of BEFV and studied protein synthesis in BEFV-infected cells.…”
