Mannan‐Binding Lectin Deficiency is Associated with Unexplained Recurrent Miscarriage

Abstract: Mannan-binding lectin (MBL) is a plasma protein which, upon binding to microbial carbohydrate structures, elicits activation of the complement system. The level of MBL is genetically determined. It has been reported that the frequency of low plasma levels of MBL is increased in patients with unexplained recurrent miscarriages (RM). In the present study plasma MBL levels were determined in 146 Danish women with RM and 41 of their husbands together with 49 Scottish RM women and 41 of their husbands. In both coun… Show more

“…More detailed analysis of the MBL gene revealed seven distinct MBL haplotypes in humans, four of which (LYPB, LYQC, HYPD, and LXPA) dictate low serum levels (45,47). MBL deficiency, defined as Ͻ100 ng of MBL per millileter in plasma, is very common in some ethnicities from 15% of the Caucasian population to as high as 30% in certain African populations (45,(47)(48)(49)(50). Compelling clinical studies have suggested that MBL deficiency was associated with severe infections following chemotherapy, postoperative, and after stem cell transplantations (51)(52)(53)(54)(55).…”

“…The samples were added to the wells, and bound H-ficolin was detected as above. Inhibition of MBL binding was tested in the same manner on the mannan-coated wells, and bound MBL was quantified using europium-labeled monoclonal anti-MBL antibodies as described previously (38,39). (24) as well as recombinant M-ficolin (produced in CHO cells and kindly provided by Anne Trommelholt Jepsen, University of Southern Denmark, Denmark) were added to the wells in 100 l of TBS/ tw/Ca 2ϩ and incubated overnight at 4°C.…”

“…An ELISA of mannan-binding activity was used to detect functional oligomers of the MBL protein in serum. 30 The assay was performed in the Department of Microbiology and Immunology, University of Melbourne, without reference to the participant's genotype. Rabbit antihuman MBL serum was obtained from Drs Kazue Takahashi and Alan Ezekowitz, Department of Pediatrics, Massachusetts General Hospital, Boston, USA.…”
