Malvidin Protects WI-38 Human Fibroblast Cells Against Stress-induced Premature Senescence

Abstract: Background:Malvidin is one of the most abundant components in red wines and black rice. The effects of malvidin on aging and lifespan under oxidative stress have not been fully understood. This study focused on the anti-aging effect of malvidin on stress-induced premature senescence (SIPS) in WI-38 human lung-derived diploid fibroblasts. Methods:In order to determine the viability of WI-38 cells, MTT assay was conducted, and malondialdehyde level was determined using thiobarbituric acid-reactive substance assay… Show more

“…This result was similar to the anthocyanins components that were reported in blueberries [69], red rice [70] and grapes [71]. Malvidin has been shown to protect against cardiovascular disease [72], and attenuate H 2 O 2 -induced oxidative stress [73] and inflammation [74], demonstrating that this strain has great potential for medical application. Except for malvidin, other content of anthocyanin components in the purple pigments were not exactly the same as that of anthocyanin components reported in above-mentioned plants.…”

“…The alkaloids fraction ( Table 3) consisted mainly of indicine-N-oxide (52.1%), ellipcine (8.1%), crinane-3α-ol (4.5%), augustamine (4.4%), 1β,2βepoxyambelline (3.8%), cinchonine (3.8%), 13α-hydrorhombifoline (3.5%), dihydro-oxo-demethoxyhaemanthamine (3.0%), oxoassoamine (3.0%), caffeine (2.3%), augustifoline (2.1%), 9-octadecinamide (2.0%), theobromine (2.0%), thalicarpin (1.7%), lupanine (1.4%), and crinamidine (1.3%). Malvidin possesses antioxidant, anti-inflammatory, cardioprotective, antihypertensive, and antitumor properties (Huang, Zhu, Li, Sui, & Min, 2016;Quintieri et al, 2013;Seo et al, 2016). β-Carotene is an antioxidant and antiosteoporotic agent (Rao & Rao, 2007), and a major precursor of vitamin A (Agea et al, 2014).…”

Therefore, a great number of potentially active molecules present in the leaves of Talinum triangulare, as well as the multifunctional properties these compounds, make Talinum triangulare a good source of healthpromoting substances.…”

“…Lack of any balance between ROS synthesis and antioxidants is referred to as oxidative stress 7 . Inflammatory factors such as NF‐κβ and its associated genes, namely, inducible nitric oxide synthase (iNOS) and COX‐2, can be triggered by oxidative stress, which results in the aging process caused by the pervasive inflammation‐related responses 8 . By scavenging free radicals such as hydroxyl radicals and superoxide anions, antioxidants can reduce the degree of damage caused by ROS, thus helping to prevent organisms' aging 4 .…”
