Maize DNA Polymerase 2 is a Phosphoprotein with Increasing Activity During Germination

Abstract: DNA replication is a late event during maize germination and DNA polymerase activity increases as germination proceeds. A replicative a-type DNA polymerase has been purified from maize seeds (DNA polymerase 2) and has been shown to be a multisubunit complex [Coello, P., Rodriguez, R., Garcia, E. Phosphate incorporated into the different polypeptides in the 11 -14-h period remained stable for at least the next 10 h (to 24 h of germination), the period of maximal enzyme activity. However, DNA polymerase 2 proces… Show more

“…A certain amount of PCNA protein is present in dry seeds, as we have reported for other cell cycle-related proteins and activities in maize dry seeds, such as replicative-type DNA polymerases and DNA primase (Coello andVázquez-Ramos 1995, García et al 1997), DNA ligase , putative cyclin B, cyclin D, Cdk4 kinase, E2F and p53 proteins (Cruz- García et al 1998) and p34 cdc2 kinase (Herrera-Teigeiro et al 1999). It would appear that during the seed development program, there might be a decision to store a potential amount of cell cycle proteins that will be required and essential for the germination process.…”

“…merases which have been identified in several plant species such as wheat [39][40][41][42], maize [43][44][45][46] and pea [47]. A challenge for the future is to identify which DNA polymerase activities act during this stage of the geminiviral DNA replication cycle.…”

“…DNA pol 1 has a native molecular mass of 350 kDa, prefers poly‐dA/oligo dT to activated DNA or poly‐rA/oligo dT and its activity is stimulated by maize PCNA (García et al 1997). DNA pol 2 has a native molecular mass around 400 kDa, its activity increases importantly during maize germination (Coello et al 1992), appears to be regulated by phosphorylation and it is a low processivity enzyme (Coello and Vázquez‐Ramos 1995a, 1995b) and contains a strongly associated DNA primase activity (García et al 2002).…”
