Maine Caucasian Population DNA Database Using Twelve Short Tandem Repeat Loci

Abstract: A population study of Caucasians residing in Maine was conducted using the AmpF1STR™ Profiler PCR Amplification Kit and the AmpF1STR™ Profiler Plus PCR Amplification Kit (Applied Biosystems Division (ABD) of Perkin Elmer, Foster City, CA). The kits contain the reagents necessary to amplify 12 different STR loci and the gender marker Amelogenin using two multiplex PCR, each containing nine STR loci. Thus, there is an overlap of six STR loci. The 12 STR loci are TH01, TPOX, CSF1PO, D3S1358, vWA, FGA, D8S1179, D2… Show more

“…This kit uses multiplex PCR amplification of nine short tandem repeat loci (D3S1358, VWA, FGA, D5S818, D13S317, D7S820, D8S1179, D21S11 and D18S51) plus the gender marker Amelogenin that allows quantification of the percentage of donor and recipient DNA in the sample studied. Because of the large number of loci that can be analysed, the power of discrimination is greater than 99% in various caucasian populations (Entrala et al , 1998; Kupferschmid et al , 1999).…”

“…In the last decade, DNA parentage tests have been widely applied to forensic cases Kupferschmid et al, 1999). Thirteen autosomal STRs of the combined DNA index system (CODIS), D5S818, FGA, D3S1358, TH01, D13S317, D16S539, D8S1179, D21S11, D7S820, CSF1PO, vWA, TPOX, and D18S51, are the basic loci of choice for forensic use.…”

“…Therefore we performed co-amplification of 12 sensitive and reliable STR loci with a combined probability of exclusion of > 0.99999999 (Hantschel et al 1999;Kupferschmid et al 1999;Neuhuber et al 1999). Staining with HE prior to laser microdissection obviously did not affect PCR amplification (Banaschak et al 2000).…”
