Mycotoxin contamination of food crops is a global challenge due to their unpredictable occurrence and severe adverse health effects on humans. Therefore, it is of great importance to develop effective tools to prevent the accumulation of mycotoxins through the food chain. The use of magnetic nanoparticle (MNP)‐assisted biosensors for detecting mycotoxin in complex foodstuffs has garnered great interest due to the significantly enhanced sensitivity and accuracy. Within such a context, this review includes the fundamentals and recent advances (2020–2023) in the area of mycotoxin monitoring in food matrices using MNP‐based aptasensors and immunosensors. In this review, we start by providing a comprehensive introduction to the design of immunosensors (natural antibody or nanobody, random or site‐oriented immobilization) and aptasensors (techniques for aptamer selection, characterization, and truncation). Meanwhile, special attention is paid to the multifunctionalities of MNPs (recoverable adsorbent, versatile carrier, and signal indicator) in preparing mycotoxin‐specific biosensors. Further, the contribution of MNPs to the multiplexing determination of various mycotoxins is summarized. Finally, challenges and future perspectives for the practical applications of MNP‐assisted biosensors are also discussed. The progress and updates of MNP‐based biosensors shown in this review are expected to offer readers valuable insights about the design of MNP‐based tools for the effective detection of mycotoxins in practical applications.

Due to the high toxicity of aflatoxin B 1 and its risks to human health, we developed a click reaction-mediated automated fluorescent immunosensor (CAFI) for sensitive detection of aflatoxin B 1 based on the Cu(I)-catalyzed click reaction. With its large specific surface area, a copper-based metal−organic framework (Cu-MOF) was synthesized to adsorb and enrich the copper ion (Cu(II)) and then load the complete antigen (BSA-AFB 1 ). After the immunoreaction, Cu(II) inside the Cu-MOF-Antigen conjugate would be reduced to Cu(I) in the presence of sodium ascorbate, which triggered the click reaction between the fluorescent donor-modified DNA and the receptor-modified complementary DNA to lead to a fluorescence signal readout. The whole reaction steps were finished by the self-developed automated immunoreaction device. This CAFI method showed a limit of detection (LOD) of 0.48 pg/mL as well as a 670-fold enhancement in sensitivity compared to conventional ELISA, revealing its great potential in practical applications and automated detection.

The rapid, simultaneous, and accurate identification of multiple non‐nucleic acid targets in clinical or food samples at room temperature is essential for public health. Argonautes (Agos) are guided, programmable, target‐activated, next‐generation nucleic acid endonucleases that could realize one‐pot and multiplexed detection using a single enzyme, which cannot be achieved with CRISPR/Cas. However, currently reported thermophilic Ago‐based multi‐detection sensors are mainly employed in the detection of nucleic acids. Herein, this work proposes a Mesophilic Argonaute Report‐based single millimeter Polystyrene Sphere (MARPS) multiplex detection platform for the simultaneous analysis of non‐nucleic acid targets. The aptamer is utilized as the recognition element, and a single millimeter‐sized polystyrene sphere (PSmm) with a large concentration of guide DNA on the surface served as the microreactor. These are combined with precise Clostridium butyricum Ago (CbAgo) cleavage and exonuclease I (Exo I) signal amplification to achieve the efficient and sensitive recognition of non‐nucleic acid targets, such as mycotoxins (<60 pg mL−1) and pathogenic bacteria (<102 cfu mL−1). The novel MARPS platform is the first to use mesophilic Agos for the multiplex detection of non‐nucleic acid targets, overcoming the limitations of CRISPR/Cas in this regard and representing a major advancement in non‐nucleic acid target detection using a gene‐editing‐based system.
