MAGEA1 interacts with FBXW7 and regulates ubiquitin ligase-mediated turnover of NICD1 in breast and ovarian cancer cells

Abstract: Melanoma-associated antigen A1 (MAGEA1) is member of the MAGE gene family that is expressed in male germ line cells and placenta under normal physiological conditions. Although MAGEA1's expression levels have been evaluated as one of the cancer testis (CT) antigens for immunotherapy in melanoma and several other cancers, its functional role and signaling mechanisms are largely unknown. In this study, we examined the functional involvement and signaling mechanisms of MAGEA1 in breast and ovarian cancer cells. I… Show more

“…27 Targeting melanoma antigen A (MAGEA1), FBXW7 is involved in the ubiquitination and degradation of the Notch1 intracellular domain (NICD1), resulting in migration and proliferation of breast and ovarian cancercells. 28 facilitates gefitinib resistance by strengthening epithelialmesenchymal transition. 29 Increased fbxw7 and decreased skp2, switch stem cells from mitotic division to quiescence in lung adenocarcinoma, by regulating c-myc and p27.…”

“…It has been reported that MAGEA1 can inhibit the proliferation and migration of MCF-7 and MDA-MB-231 breast cancer cell lines (14). In addition, MAGEA1-A3 and A12 have been investigated in the early detection of breast cancer (15).…”

“…In addition, it has been reported that FBXW7 functions as a tumor suppressor, inhibits breast cancer growth and promotes cell apoptosis by targeting metadherin for degradation (41). A previous study reported that melanoma-associated antigen 1 interacts with FBXW7 to regulate the ubiquitin ligase-mediated turnover of Notch1 intracellular domain 1 in breast and ovarian cancer cells (42). FBXW7 upregulation also suppresses metastasis and epithelial mesenchymal transition in RCC (43).…”
