Macambira plant (Bromelia laciniosa Mart. Ex Shult. & Schult. f.) is a species belonging to the family Bromeliaceae, which is distributed widely in the hinterlands of northeastern Brazil, from Bahia to Piauí states. The plants of this species contain an enzyme complex named bromelain, which has great potential for therapeutic applications. In the present study, bromelain derived from Bromelia laciniosa leaves was assessed for its thermodynamic properties and in vitro cytotoxic and healing activities. Bromelain was extracted from the leaves and partitioned using ethanol. The partially purified bromelain was then characterized to determine the optimum pH and temperature and the kinetic and thermodynamic parameters. The partially purified bromelain derived from macambira leaves exhibited its highest activity at 47°C and pH 7, when the Km and Vmax values of 2.26 µM and 2.083 U/mL, respectively. The half-life of this bromelain at 47°C was 5.78 h. At 47°C, the Gibbs's free energy, enthalpy, and entropy values reached 89, 53.54, and –0.11 kJ/mol K, respectively. In the SDS-PAGE analysis, bromelain appeared as the main protein band, with an estimated molecular mass of 33 kDa. The in vitro cytotoxic activity and wound healing effects were determined using the MTT assay and Scratch assay, respectively. These assays were performed on the non-neoplastic cell lines L-929 and PBMC and the human tumor cell lines MCF-7, HCT-116, and NCI-H292. Bromelain exhibited a positive healing activity with a change rate similar to that observed for the control. In addition, bromelain exhibited cytotoxic activity against NCI-H292 tumor cells and chemopreventive potential. These results indicate that bromelain derived from macambira leaves may be utilized as a potential ingredient in pharmaceutical formulations, for various healing and chemopreventive purposes and as an alternative to the bromelain extracted from pineapple. In addition, the biochemical characteristics of this bromelain could find application in the food industry. Future studies should, therefore, focus on deciphering the action mechanisms underlying the activities of bromelain observed in the present study to further confirm and enhance its application potential for therapeutic purposes.

ABSTRAKBonggol belum dimanfaatkan secara optimal, padahal bagian bonggol mengandung beberapa komponen aktif salah satunya adalah enzim bromelin. Ezim bromelin bersifat sebagai antibakteri. Tujuan penelitian ini untuk mengetahui efektivitas daya hambat ekstrak etanol 96% bonggol nanas (Ananas comosus L) terhadap perumbuhan bakteri Staphylococcus aureus. Penelitian ini merupakan penelitian eksperimental menggunakan desain post-test only control group design. Ekstrak etanol 96% bonggol nanas (Ananas comosus L) dilaukan ekstra dengan metode maserasi Bakteri Staphycoccus aureus diambil dari media Manitol salt agar. Hasil penelitian ini menunjukkan ekstrak bonggol nanas pada konsentrasi 50%-100% dengan diulang sebanyak 3 replikasi memiliki daya hambat terhadap pertumbuhan bakteri Staphylococcus aureus. Berdasarkan uji One-Way ANOVA dengan taraf kepercayaan 95% menunjukkan terdapat perbedaan yang bermakna (p<0,05) antara berbagai konsentrasi, sehingga dilanjutkan dengan uji LSD untuk mengetahui adanya perbedaan pada masing-masing perlakuan. Hasil uji LSD menunjukkan bahwa kosentrasi 50% berbeda nyata dengan konsentrasi 70%, 80%, 90% dan 100%, sedangkan control positif tidak berbeda nyata dengan konsentrasi 50% dan 60%. Kesimpulan penelitian ini bahwa ekstrak etanol 96% bonggol nanas (Ananas comosus L) berpengaruh positif dalam menghambat dan membunuh bakteri Staphylococcus aureus dan konsentrasi paling optimal pada 70%.Kata kunci: Ekstrak etanol 96% bonggol nanas, bacitrasin, Staphylococcus aureus, zona hambat antibakteriABSTRACTPineaplle cobs have not been utilized optimally, whereas the cob section contains several active components one of which is the enzyme bromelin. Bromelin is an antibacterial agent. The purpose of this research was to determine the effectiveness of inhibitory of ethanol extract 96% pineapple cobs (Ananas comosus L) to Staphylococcus aureus bacteria growth. This research is an experimental using post-test design only control group design. Pineaplle cobs extract 96% by maceration method, Staphycoccus aureus bacteria taken from Mantol salt agar medium. The results of this study showed the pineapple cobs extrac concentrations of 50% -100% with replication as any 3 replications have an inhibitory to the growth of Staphylococcus aureus bacteria. Based on One-Way ANOVA test with 95% confidence level there was a significant difference (p <0,05) between various concentrations, so it was continued with LSD test to know the difference in each treatment. LSD test results showed that 50% concentration was significantly different with concentrations of 70%, 80%, 90% and 100%, while the positive control did not differ significantly with concentrations of 50%. The conclusion of this research is that pineapple cobs(Ananas comosus L) has positive effect in inhibitied and killing Staphylococcus aureus bacteria and optimal concentration 70%. B ACKGROUND:Alpinia galanga (A. galanga) was reported as a potential medicinal source due to its wide effect. A. galanga rhizome crude extract (ARCE) was reported to have high cytotoxic effect in cancer cells, but low in normal cells. However half maximal inhibitory concentration (IC 50 ) of ARCE is not clearly known yet. Hence, current study was conducted to investigate the IC 50 of ARCE in normal standard fibroblast cell line, NIH-3T3 cells. METHODS:Rhizomes of A. galanga were collected, peeled, dried, milled and weighed. Extraction was performed using maceration method, then filtered and evaporated. ARCE with various concentrations were applied in NIH-3T3 cells for 24 or 48 hours. Cells were documented and counted with 3-(4,5-dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium bromide (MTT) assay. RESULTS:Five hundreds grams of simplicia were macerated with ethanol and evaporated, 1 mg/mL crude extract with total volume of 114 mL was obtained. By addition of ARCE in NIH-3T3 cell culture, number of NIH-3T3 cells were shown less when treated with higher concentration of ARCE. Cell numbers of 0, 3.125, 6.25, 12.5, 25 and 50% ARCE treatment for 24 hours are 11, 531, 11,352, 10,920, 10,365, 9,471, 8,360, respectively, meanwhile for 48 hours are 13,219, 12,686, 12,278, 11,390, 10,279, 8,390, respectively. CONCLUSION: IC 50 of ARCE in 24 hours treatment was 620.5 mg/mL, while in 48 hours treatment was 666.6 mg/ mL. Hence, ARCE is suggested to have low cytotoxic effect in NIH-3T3 cells. KEYWORDS:
