Macelignan protects HepG2 cells against tert‐butylhydroperoxide‐induced oxidative damage

Abstract: In this study, we investigated the protective effect of macelignan, isolated from Myristica fragrans Houtt. (nutmeg) against tert-butylhydroperoxide (t-BHP)-induced cytotoxicity in a human hepatoma cell line, HepG2. The tetrazolium dye colorimetric test (MTT test) and lactate dehydrogenase (LDH) assay were used to monitor cell viability and necrosis, respectively. Lipid peroxidation [malondialdehyde (MDA) formation] was estimated by the fluorometric method. Intracellular reactive oxygen species (ROS) formation… Show more

“…Many compounds and derivatives such as a-tocopherol, niacinamide, oxyresveratrol, ascorbic acid, and lipoic acid can inhibit melanin synthesis. [34][35][36][37][38] Sohn et al 21) reported that macelig- nan had high free radical-scavenging activity and inhibitory effects on lipid peroxidation induced by tert-butylhydroperoxide. Therefore its melanogenesis-inhibitory activity is thought to be largely due to its antioxidant effects as well as direct inhibition of tyrosinase.…”

“…[13][14][15] Macelignan isolated from M. fragrans belongs to the lignan constituents, 16) and has been reported to have a broad spectrum of biological activities such as antioxidant, antiinflammatory, anticarinogenic, and hepatoprotective effects and cause alteration in hepatic enzyme activities. [17][18][19][20][21] However, the effects of macelignan on melanogenesis have not been examined so far. In this study, we evaluated the inhibitory activity of macelignan on melanin biosynthesis and melanogenic protein expression using melan-a murine melanocytes.…”

“…Therefore, understanding the role played by antioxidants agents during drug-mediated toxicity is important to determine if they have a protective effect against oxidative stress induced by reactive intermediates (41,42).…”

“…(nutmeg), exerts anti-oxidant, anti-inflammatory, anti-carcinogenic, anti-diabetic, and hepatoprotective effects. [15][16][17] In addition, macelignan has been reported to inhibit melanin biosynthesis in murine melan-a melanocytes. 18) However, the effects of macelignan on melanosome transfer have not been determined.…”
