Lymphocyte antigens in sheep: linkage to the MHC class II DRB1 gene

Abstract: In this work a typing battery of sera was developed to test lymphocyte antigens in sheep. Eight antigens were detected in a Latxa sheep sample. The serological determination of these antigens is described. As some of the detected antigens segregated in close linkage with class II DRB1 SSCP patterns in two half-sib families, we can conclude that they are coded by genes located in the MHC. Gene frequencies were very similar in Latxa Mutur Gorria and Latxa Mutur Beltza, the two varieties of the Latxa breed. Altho… Show more

“…Heterozygosity was significantly higher (96%; P < 0.05, t-test) among the Valachian sheep as described in Table1. Different SSCP patterns of DRB1 gene have been reported earlier (Kostia et al 1998;Jugo and Vicario 2000;Jugo and Vicario 2001). The frequency of the k genogroup was the highest (28%) followed by genogroup e (21%).…”

“…The animals were serologically typed for lymphocyte antigens by a standard two‐stage lymphocyte microcytotoxicity assay (Terasaki & McClelland, 1964), using a battery of locally raised alloimmune antisera according to Jugo & Vicario (2001).…”

“…Extensive polymorphism for ovine lymphocyte antigen (OLA) class I antigens has been demonstrated by serological methods, and reports from different groups describe up to 31 provisional specificities, with indications that these are controlled by codominant alleles at two or more autosomal loci (reviewed in Spooner et al ., 1988). Recently, more laboratories are producing valuable reagents (Garrido et al ., 1995; Stear et al ., 1996; Jugo & Vicario, 2001).…”

“…MHC Class II Ovar- DRB 1 was chosen as the immune response gene in this study because it is highly polymorphic, transcribed, and there are over 100 different DRB 1 alleles reported in Genbank based upon either restriction fragment length polymorphisms (RFLP) or the deduced amino acid sequence for the β1 domain encoded by exon 2 (Dutia et al , 1994; Schwaiger et al , 1996; Jugo & Vicario, 2001; Konnai et al , 2003; Herrmann et al , 2005). Others have shown specific MHC- DRB 1 alleles associate with resistance and/or less severe clinical signs in human hydatidosis (Gottstein et al , 1996; Li et al , 2003).…”
