Lycorine hydrochloride inhibits metastatic melanoma cell‐dominant vasculogenic mimicry

Abstract: Summary Melanoma cells actively participate in tumor angiogenesis and vasculogenic mimicry. However, anti‐angiogenic therapy in patients with melanoma has not shown a significant survival gain. Thus, new anti‐melanoma angiogenic and vasculogenic drugs are highly desired. Using the metastatic melanoma cell line C8161 as a model, we explored melanoma vasculogenic inhibitors and found that lycorine hydrochloride (LH) effectively suppressed C8161 cell‐dominant formation of capillary‐like tubes in vitro and generat… Show more

“…In addition, not all tumors with evidence of VM expressed DSG2. It is likely that DSG2 is one of several cadherins which can contribute to VM, and indeed, previous studies have implicated VE-cadherin in this process [47, 48]. Furthermore, other members of the desmosomal cadherin family were expressed at low level in a small number of TCGA tumors, and DSG1, DSC2 and DSC3 were all significantly enriched in the DSG2 -high tumor subset.…”

“…However, lycorine was found to induce apoptosis as well as arrest cell cycle of HL-60, KM3, K562, Hey1B cells in different phases [7,[14][15][16]. Recently, the inhibitory effect on vasculogenic mimicry of melanoma cells [17] and suppression of neovascularization of ovarian cancer Hey1B cells by lycorine were reported [16]. Many factors thus seem involved in the anticancer properties of lycorine and a clear mechanism remains to be determined.…”

“…It is not completely clear whether VM reflects the plasticity of differentiated melanoma cells or transdifferentiation of melanoma CSC. Nonetheless, many different pharmacologic interventions have been reported to inhibit VM in various cancers [75][76][77][78][79][80][81]. Among these, thalidomide, endostatin, and lycorine hydrochloride were shown to inhibit VM specifically in melanomas [80,81].…”

Other compounds known to inhibit tumor VM either in vitro or in an in vivo mouse model, include: isoxanthohumol, endostar, zoledronic acid, curcumin, and a group of cellpermeable, phosphatase-stable phosphopeptide mimetic pro-drugs targeted to the SH2 domain of Stat3 [75][76][77][78][79].…”
